稻苗草榨汁對抑制發炎反應的功能研究

Study of the Anti-inflammatory Effect of Rice Shoot Juice

摘要


本研究以巨噬細胞株、初代腹腔巨噬細胞和動物模式,探討稻苗草對抗發炎的影響。先以細胞培養模式,評估台中秈17、台中糯70、台農67等三種稻苗草榨汁,對發炎介質分泌的影響,再以動物試驗,探討管餵動物稻苗草後,腹腔巨噬細胞分泌發炎介質之變化。結果顯示,稻苗草與裂殖素活化的巨噬細胞共同培養後,三種稻苗草皆能抑制活化之RAW264.7和初代腹腔巨噬細胞分泌NO (nitric oxide)與interleukin (IL)-6的作用。其中台中秈17同時又能降低prostaglandin E2 (PGE2)與tumor necrosis factor (TNF)-α的分泌,並可促進脾臟細胞IL-2的分泌和脾臟細胞增生能力的趨勢。進一步以台中秈17進行動物試驗,分為控制組、台中秈17低劑量組(3毫克/隻/天)與台中秈17高劑量組(10毫克/隻/天)三組。管餵BALB/c雌鼠六週後犧牲,取腹腔巨噬細胞以lipopolysaccharide與interferon-γ(LPS/IFN-γ)活化後,分析發炎介質。結果顯示管餵BALB/c雌鼠低劑量台中秈17榨汁,顯著降低腹腔巨噬細胞分泌IL-6與TNF-α之能力。因此,本研究結果顯示,以RAW264.7和初代腹腔巨噬細胞可作為初步篩選抗發炎潛力食材之細胞模式,且以動物餵食劑量估算,在動物試驗可獲一致結果。因此,本研究採用的試驗細胞與組織細胞之比例推估可作為餵食動物劑量之參考。同時,台中秈17稻苗草具降低發炎介質之能力,對於發炎相關疾病的影響或減緩發炎反應,值得進一步研究。

To exploit healthy food from herbaceous plants in Taiwan, we investigated whether the anti-inflammatory effect of target samples could be screened by using RAW264.7 cell and primary peritoneal macrophage cell culture models. First, the effect of rice shoot juice, from three different rice species TCS17, TCW70 and TN67, on inflammatory mediators secretions by these cells was evaluated. Sequentially, animal study was also performed to confirm the results. The splenocyte proliferation assay was also measured using (superscript 3)H-thymidine incorporation method. The results showed that three kinds of rice shoot juice reduced NO and IL-6 secretion in LPS/IFN-γ-activated RAW264.7 cells and peritoneal macrophages. Moreover, TCS 17 also inhibited PGE2 and TNF-α secretion of activated macrophages, and promoted IL-2 secretion and cell proliferation of splenocytes. These results suggested TCS17 possesses the immune-regulatory potential. Therefore TCS17 was further used for tube-feeding to BALD/c female mice which were divided to the control group, low dose group (3 mg TCS17/mouse/day), and high dose group (10 mg TCS17/mouse/day). After six-weeks’ tube fed of TCS17, BALB/c mice were sacrificed for ex-vivo study. The peritoneal macrophages were stimulated with LPS/IFN-γ and the cell supematants were collected for measurement of inflammatory mediators. The results showed that TCS17 groups had lower IL-6 and TNF-a secretion compared to the control group. Therefore, RAW264.7 cell and prithary peritoneal macrophage cell culture could be as primary screening method for anti-inflammatory substance. The cell to tissue ratio for estimating the dosage of target samples in animal study from cell culture may be applicable. The anti-inflammatory effect ofTCSl7 suggested that herbaceous plants may possess anti-inflammatory function and is worthy of further study.
