本研究首先利用芝麻榨油後所得之富含醣基芝麻木酚素的芝麻粕為原料,篩選芝麻木酚素去醣基活性強的菌株,並利用體外試驗探討發酵物之乙醇粗萃物的抗氧化能力及抑制癌細胞株增生的能力。結果顯示,分離菌株Aspergillus sp. A-23具有最高的去醣基芝麻木酚素的生成能力,且相較於未發酵的芝麻粕,其可顯著提升發酵後產物的酒精粗萃物之總抗氧化能力(trolox equivalent antioxidant capacity; TEAC)、清除1,1-diphenyl-2-picrylhydrazyl(DPPH)自由基能力、脂質抗氧化能力和對銅離子誘導的LDL過氧化的抑制能力以及抑制胃癌(AGS)、子宮頸癌(Hela)、乳癌(MCF-7)、肝癌(Hep G2)及大腸癌(Caco-2)的細胞增生。試驗結果得知,經分離菌株Aspergillus sp. A-23發酵後的芝麻粕發酵物之乙醇粗萃出物具有減少氧化壓力及抑制癌細胞增生的效應。
In this study, we first utilized sesame meal after extracting sesame oil rich in sesaminol glycosides to screen organisms for high sesaminol deglycoside activity and investigated the antioxidant properties and anticancer capacity in vitro. Strain Aspergillus sp. A-23, having the highest sesaminol deglycoside activity, was isolated and applied for fermenting sesame meal. The ethanol extracts of fermented sesame meal significantly elevated trolox equivalent antioxidant capacity (TEAC), DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging ability, antioxidant effect on liposome oxidation and inhibition ability of Cu(superscript 2+)-induced lipid peroxidation in LDL. Meanwhile, it also in creased the antiproliferative effects on AGS、Hela、MCF-7、HepG2 and Caco-2 cells in vitro, as compared to unfermented sesame meal. These results demonstrated that ethanol extracts of sesame meal fermented with Aspergillus sp. A-23 has the ability to decrease oxidative pressure and antiproliferative effects on carcinoma cells.
