明日葉之一般組成及抗氧化性質

Proximate Composition and Antioxidative Properties of Angelica keiskei Koidzumi

摘要


本研究探討明日葉(Angelica keiskei Koidzumi)經冷凍乾燥及40℃熱風乾燥後,其一般組成、礦物質含量及其類超氧歧化酶活性之差異性,評估最適之乾燥方式。再經溶劑(甲醇、水、正己烷及乙酸乙酯)萃取,比較其萃取率並選取最適溶劑後,配合不同萃取時間(1、3、5、8、12及24小時)進行抗氧化能力及抗氧化成分分析。結果顯示,冷凍乾燥葉部類超氧歧化酶活性(102.4Unit/g)較莖部(42.5Unit/g)及其他熱風乾燥者(葉58.9及莖55.8Unit/g)為高,顯示熱風乾燥會影響類超氧歧化酵素活性。抗氧化試驗方面,甲醇為最佳萃取溶劑,萃取時間與萃取率呈相關性,然萃取時間並不會對抗氧化能力造成差異;葉部之抗氧化能力於各試驗項目(清除DPPH自由基能力、還原力、總抗氧化能力及抑制脂質過氧化能力)皆優於莖部。分析其抗氧化成分,得知葉部的酚類化合物含量為莖部的5.9-6.8倍,且綠原酸(chlorogenic acid)含量佔了酚類化合物的43%。綜合以上結果可得知,冷凍乾燥為較適之乾燥方式,且以葉部具有較強之抗氧化能力及較高量的抗氧化成分。

The present study was to investigate the effects of freeze-drying and oven-drying (40℃) on the composition, mineral contents, and SOD-like activity of Angelica keiskei Koidzumi, and then determine the most suitable drying method. After the extraction with different solvents such as methanol, water, hexane, and ethyl acetate, the solvent giving the highest extraction yield was used. The antioxidative activities and the active components content of the extracts obtained with different extraction time (1, 3, 5, 8, 12, and 24 h) were determined. The methanol extract was found to have the highest antioxidative activities. A high correlation was found between the extraction time and extraction yield, while the extraction time did not affect the antioxidative activities. The results showed that the SOD-like activity of the freeze-dried leaves (102.4 Unit/g) was higher than that of the freeze-dried stem (42.5 Unit/g) as well as those of oven-dried materials (58.9 Unit/g for leaves and 55.8 Unit/g for stem). The results demonstrated that the SOD-like activity was affected by oven-drying. The antioxidative activities of the extracts from leaves (e.g. DPPH scavenging capacity, reducing power, total antioxidative activity, and lipid peroxidation inhibitory activity) were higher than those from stem. Chemical analyses showed that the amounts of phenolic compounds in the extracts from leaves were 5.9-6.8 times higher than those of stem, while the chlorogenic acid accounted for 43% of the total content of phenolic compounds. The above results showed that freeze-drying was the suitable drying method, while the antioxidative activities and active compound contents of the extracts from leaves were higher than those from stem.
