利用HPLC分析食藥用菇類中腺苷之方法研究

Determination of Adenosine in Edible and Medicinal Mushrooms by High Performance Liquid Chromatography

摘要


腺苷為一影響人體多種生理活動之成分,且為食藥用菇類重要之機能性指標之一,本研究之目的係以高效能液相層析儀配備紫外線偵檢器,建立一快速且靈敏之分析方法,以測定食藥用菇類中腺苷含量,內容包括不同試驗條件,如移動相組成、萃取方式、萃取溶劑及萃取時間之影響探討,在選定的試驗條件下,該方法之量測不確定度為225.0±11.9μg/g,儀器偵測極限及方法偵測極限則分別為 0.2 μg/g 及10.0 μg/g。再者,藉由標準品添加之測定結果來確認本方法之準確度,其回收率可達96-105%,變異係數(CVs)均在3.5%以下。另外,將本方法應用於不同種類菇類之腺苷含量測定,結果證明本方法簡易又快速,适合用於大量樣品之例行分析。

Adenosine is a molecule that influences various physiological systems in human body, and is one of the important functional components in edible and medicinal mushrooms. The purpose of this study is to establish a rapid and sensitive method, high-performance liquid chromatography with UV detector, to determine adenosine content in edible and medicinal mushrooms. Furthermore, the effects of various experimental conditions including mobile phase composition, extraction system, extraction time and extraction solvent were also optimized in this work. Under specific conditions, the measurement uncertainty of the proposed method is 225.0 ± 11.9 μg/g, along with the instrument and method detection limit are 0.2 μg/g and 10.0 μg/g, respectively. The recovery by this method ranged from 96 to 105%, with coefficients of variation (CVs) less than 3.5%. The proposed method was subsequently applied to quantify adenosine content in different kinds of mushroom samples. The results showed its suitability for adenosine determination in edible and medicinal mushrooms, especially for routine analysis.
