The Effects of Cholic Acid and Cyprinol Sulfate on Liver Damage in Rats Induced by Acetaminophen

膽酸和鯉魚膽醇硫酸塩對止痛藥乙醯胺酚引起大鼠肝傷害之作用

摘要


為證實傳統中藥動物膽汁補肝之功能,本研究以部分大鼠每天腹腔注射止痛藥乙醯胺酚(acetaminophen)150mg/kg共四天,誘發大鼠產生肝病變後,將肝病變大鼠(乙醯胺酚處理)群和未病變大鼠(乙醯胺酚未處理)群,再各分四組,每組六隻大鼠,每組各隻大鼠每三天各口服生理食鹽水(對照組),10mg膽酸(cholicacid)、20mg膽酸和10mg鯉魚膽醇硫酸塩。經29天實驗期間後,乙醯胺酚處理群之大鼠血漿中之AST、ALT和ALP生化指標均顯著較乙醯胺酚未處理群者上升。但是29天後,肝體比和腎體比,血液中紅血球濃度、紅血素、血體積比,血漿中之AST、ALT、ALP、BUN和creatinine濃度,肝臟中glutathine和thibarbituricacid-relativesubstance量,在兩群中並無差量,顯示膽酸和鯉魚膽醇硫酸塩並無減緩乙醯胺酚導致肝病變之作用。

To investigate the effects of bile acids and bile alcohols on liver function, male Wistar rats were used as experimental animals in this study. The rats were divided into two sections: the control section (with no acetaminophen being administered) and the experimental section (with acetaminophen being administered). Liver damage was induced in the experimental section by administering acetaminophen to the rats daily (150 mg/kg daily, i.p.), for 4 days. Each of these two sections (experimental and control) was divided once more into four groups of six rats each. Each set of six rats, within each of the two sections, was treated orally, at 3-day intervals, with either saline solution, 10 mg of cholic acid, 20 mg of cholic acid or 10 mg of cyprinol sulfate, respectively. During the 29-day experimental period, the rats were allowed food and drinking water ad libitum. After the acetaminophen had been administered, the AST, ALT, and ALP activity in the rats’ plasma increased significantly. After 29 days, however, the relative ratios of both liver and kidney weight to body weight, concentrations of RBC, hemoglobin and hematocrit in the blood, levels of AST, ALT, ALP, BUN and creatinine in the plasma, as well as the levels of glutathione and thiobarbituric acid-reactive substances (TBARS) in the livers of the rats, did not differ significantly between the control rats and the experimental rats. This indicated that cholic acid and cyprinol sulfate did not ameliorate the damage induced by acetaminophen in the liver function of rats.
