以液相層析串聯質譜儀檢測雞蛋及鮮乳中殘留之離子攜帶型抗生素

Determination of Five Polyether Ionophore Antibiotics in Egg and Fresh Milk by Liquid Chromatography-Electrospray Tandem Mass Spectrometry

摘要


本試驗方法使用液相層析串聯質譜儀(liquid chromatography/tandem mass spectrometry, LC/MS/MS),以電灑游離正離子法,進行雞蛋及鮮乳中五種離子攜帶型抗生素之檢測及方法開發。試驗步驟為取全蛋及鮮乳加入無水硫酸鈉,混合均勻直到呈現小顆粒球狀,以乙腈萃取後,經氮氣濃縮後,選用XTerraTM Phenyl之層析管柱,搭配0.1%甲酸水溶液與乙腈進行LC/MS/MS分析。此方法之雞蛋及鮮乳平均回收率介於90.4~95.0%及95.0~104.7%間,檢測限量介於0.5~1.5 ppb間。本方法提供了快速、有效的檢驗方法,可減少人力及試驗耗費,增加檢驗的時效性。

A LC/MS/MS method for the simultaneous detection and confirmation of five polyether ionophore antibiotics in egg and fresh milk was developed and validated. The analytes were extracted and injected into the LC-ESI-MS/MS system on a C18 column in gradient mode and an isocratic elution was performed. Results showed good repeatability, and recoveries for the five polyether ionophores used in egg and in milk averaged 90.5~95.0% and 95.0~104.7%, respectively. This method was suitable to confirm the presence of ionophore antibiotic residues at concentrations of 0.5~1.5 ppb in egg and fresh milk, respectively.
