白腎豆萃取物對攝食高脂飲食大鼠醣類及脂質代謝之影響

Effects of White Kidney Bean Extracts on Carbohydrate and Lipid Metabolism in Rats Fed a High Fat Diet

摘要


肥胖症(obesity)與其衍生之相關代謝性疾病為全球重要的公共衛生議題,白腎豆(Phaseolus vulgaris)萃取物(澱粉酶抑制劑,Phase2®)為近年來廣泛應用於體重控制之機能性食品,然而其作用機制不明。本研究以Wistar大鼠作為試驗動物,探討飲食中添加白腎豆萃取物對高脂飲食大鼠醣類及脂質代謝之影響。試驗共分為三組:(一)正常飲食組(NC)、(二)高脂飲食組(HF)、(三)高脂飲食添加5%Phase2®組(P),飼養期間採自由進食,試驗共進行五週。結果顯示,攝取含Phase2®的組別能顯著降低攝食量並減少體重增加。5%Phase2®組可顯著降低血漿胰島素及HOMA-IR值,且能降低小腸蔗糖酶及麥芽糖酶活性。高脂飲食添加Phase2®能有效降低肝臟中總膽固醇及三酸甘油酯含量。在脂肪組織方面,Phase2®可藉由提升脂肪組織hormone sensitive lipase(HSL)活性來減少脂肪囤積,並減少副睪脂肪三酸甘油酯含量,提升脂肪組織脂解速率。此外Phase2®可降低血漿IL-6、TNF-α及leptin濃度。綜合以上結果,推測白腎豆萃取物具有改善肥胖及改善相關代謝疾病之潛力。

Obesity and its derivative metabolic diseases are regarded as an important global public health issue. White kidney bean extracts (Phase 2®) is widely used in functional foods to reduce weight in recent years, but its mechanisms are still unknown. This study was designed to investigate the effects of white bean extracts on glucose and lipid metabolism in high fat-feeding rats. Male Wistar rats were randomly divided into three groups: (1) normal diet control (NC), (2) high fat diet (HF) and (3) high fat diet with 5% Phase 2® supplementation. This experiment was done for 5 weeks. As the results, Phase 2® significantly reduced food intake and body weight gain. The insulin concentration and HOMA-IR index were also reduced by 5% Phase 2® feeding. Besides, the small intestinal maltase and sucrase activities was decreased after Phase 2® treatment. Rats fed high fat diet had higher hepatic total cholesterol and triglyceride contents. However, Phase 2® supplementation ameliorates the liver lipids. It is worth to note that Phase 2® induced hormone sensitive lipase (HSL) activity, leading to decreased triglyceride content in epididymal adipose tissues. Furthermore, Phase 2® decreased plasma IL-6, TNF-α and leptin concentrations and increased the fecal total cholesterol and triglyceride contents. These results suggest that white bean extracts have the potential to improve obesity and related metabolic diseases in rats fed a high fat diet.
