以果膠與羧甲基纖維素為黏著劑對流動床造粒之粒徑效應與品質之影響

The Effect of Particle Size and Quality of Fluid Bed Granulation with Pectin and Carboxymethylcellulose as Binder

摘要


流動床造粒為一濕式造粒法,常將指標物溶入黏著劑噴入賦型劑顆粒,藉黏著包覆達到造粒之目的。黏著劑之種類、濃度、黏度是重要操作因子,本實驗以羧甲基纖維素(0.5%)、果膠液(2.5%、5.0%)、果膠顆粒(5.0%、10%)五種不同濃度、種類或添加形式為黏著劑評估造粒粒徑增長效應及其粉體品質,本實驗結果如下:以0.5% CMC、2.5、5%果膠液進行造粒評估時,5%果膠液因黏度較高其在粒徑增長最快( p < 0.05),在造粒240 min可得到193.79 μm粉粒,0.5% CMC 199.73 μm、2.5%果膠液145.32 μm;在包覆量a值項目五種黏著劑包覆率相近,差異不明確;以果膠粒造粒之粉粒體積密度、振實密度較高,流動性較其他種粉粒低。在貯存期間,不同粉粒之水分、水活性均隨貯存溫度、濕度提高,時間延長呈增加變化,尤以第一天增加較明顯。

Fluid bed granulation is a kind of wetting granulation that often dissolved indicator into binder and spray on the excipient to achieve the purpose of granulation by adhesion and coating. The type, concentration and viscosity of binder are the important operation factors. In this experiment, we used carboxymethylcelllose solution (0.5%), pectin solution (2.5%, 5.0%) and pectin particles (5.0%, 10%) with five different concentration, type and adding method, as binder to evaluate granulated particle size growth effect and its quality of powder. The experimental results were as follow: because 5% pectin solution has higher viscosity, its particle size grew fastest (p < 0.05) when using 0.5% CMC, 2.5 and 5% pectin solution to evaluate granulation. After granulating 240 min, we got the particle size (5% pectin solution, 193.79 μm, 0.5% CMC solution, 199.73 μm, 2.5% pectin solution, 145.32 μm). The encapsulated amount of the color (a value) show no significant difference between five type of binder. The powder granulated by pectin particle has higher bulk density and tapped density, but lower flow ability. As temperature, humidity and time increasing, the water content and the water activity of the different powder increased during storage. And it showed a more significant increasing in the first day.
