複頻超音波輔助萃取香菇蒂粉總多醣之研究

Study on Dual-frequency Ultrasound Assisted Extraction of Total Carbohydrate from Mushroom Stalk Powder

摘要


香菇多醣具有免疫調節等多項生理功效,本研究應用超音波輔助萃取香菇蒂粉末,探討複頻處理萃取多醣效益以提高香菇副產物之利用價值。實驗以40℃、料液比1:20 g/mL、能源密度0.025 w/g之20及35 kHz單一、組合及複合操作超音波輔助萃取30分鐘,並與靜置與傳統萃取比較,探討不同處理方式對不同粒徑香菇蒂粉末萃取總多醣含量之影響。結果顯示相同時間萃取率:複頻組合式>複頻複合式>單頻操作>傳統水萃>靜置水萃,且具有粒徑與時間效應。以經過擠壓蒸煮(M1)與超微粉處理樣品(M3)萃取30分鐘為例,在20 kHz處理中其總多醣萃取率較傳統水萃(11.0 ± 0.1 mg/mL)提高近1.68倍,相當於傳統熱水萃取約160分鐘;且其複頻組合式總多醣含量為22.9 ± 0.1 mg/mL,萃取率較傳統水萃提高2.08倍,相當於傳統熱水萃取約280分鐘,顯示超音波輔助萃取具有低溫操作與節省能源效益。

Polysaccharides come from mushroom has many physiological functions such as immune regulation. In this study, ultrasound-assisted extraction of mushroom stalk powder was used to explore the benefits of dual-frequency treatment to enhance the utilization value of mushroom by-products. The experiments were conducted at 40°C for 30 minutes with a solid-liquid ratio of 1 : 20 g/mL for static and 0.025 w/g ultrasonic assisted extraction (20 and 35 kHz single and combined operations) compared with conventional hot water extraction to investigate the effect of ultrasound on the total polysaccharide content of mushrooms stalk powder with extrusion cooking and ultrafine grinding. The results show that the dual-frequency composite > dual-frequency combination > single frequency operation > traditional water extraction > standing water extraction, and has a particle size and time effects. Taking the extraction (M1) and ultrafine powder (M3) extraction for 30 minutes as an example, the total polysaccharide extraction rate in the 20 kHz treatment is nearly 1.68 times higher than that of the traditional water extraction (11.0 ± 0.1 mg/mL), which is equivalent to traditional hot water extraction for about 160 minutes; and its dual-frequency combined polysaccharide content is 22.9 ± 0.1 mg/mL, and the extraction rate is 2.08 times higher than that of traditional water extraction, which is equivalent to about 280 minutes of traditional hot water extraction. It shows that ultrasonic assisted extraction has low temperature operation and energy saving benefits.
