Immune Inhibitory Factor in Mammalian Tissues

A protein obtained from mammalian livers capable of inhibiting mitogen-induced lymphocyte proliferation, is called ”liver-derived inhibitory protein”, or LIP. LIP was purified from mouse liver homogenates by various purification procedures, including ammonium sulfate precipitation, DEAE chromatography, hydroxylapatite chromatography, and G-150 superfine Sephadex gel filtration. The LIP was proven to be very pure, and its inhibitory activity was improved 37-fold. The LIP was identified to be arginase. LIP can inhibit both T- and B-lymphocytes. It is temperature stable up to 65℃ and pH stable from 3 to 11. LIP molecule weight is 105 kDa. Arginase is the only mammalian amino acid-degradation enzyme that can potently inhibit lymphocyte proliferation. Therefore, arginase of human origin has a great potential of therapeutical benefit in treating immunologic diseases, such as transplant rejection, systemic lupus erythematosus, etc.
