自然感染豬生殖與呼吸綜合症病毒豬隻體內病毒之分佈與定量

Distribution and Quantitation of Porcine Reproductive and Respiratory Syndrome Virus in Naturally Infected Pigs

摘要


本試驗建立反轉錄競爭性定量聚合酶連鎖反應(reverse transcription-quantitative competitive PCR, RT-qcPCR)技術,進行豬生殖與呼吸綜合症病毒(PRRSV)核酸定量,並將其應用於現場自然感染豬隻體內病毒之分佈與定量研究。應用台灣PRRSV病毒株建構target sequence及internal control質體,應用此質體進行RT-qcPCR並建立了定量病毒之標準曲線(y=4.94x + 0.18 ; R^2=0.98)。仔豬之母體移行抗體試驗結果顯示,抗體於9週齡時降至最低,此時病毒血症比率達100%,其血清中病毒之平均濃度為10^4.1 copies/mL;PRRSV自然感染豬隻,其肺臟之病毒含量最高達10^5.7-10^8.9 coples/g,血清及其他臟器如扁桃腺、鼠蹊淋巴、肺門淋巴、腸繫膜淋巴及脾臟之含量亦達10^5 copies/g以上,鼻分泌液及口水亦分別有10^5.8及10^4.8 copies/mL;消化道及尿液亦分別含有10^2.5至10^3.5 copies/mL之病毒。以上結果顯示,豬群體內移行抗體下降至最低時,即是病毒血症發生之高峰期,肺臟及淋巴系統為病毒之主要標的器官,而鼻分泌液及口水為病毒之主要感染來源。

A reverse transcription-quantitative competitive polymerase chain reaction (RT -qcPCR) was established for the quantitation of porcine reproductive and respiratory syndrome virus (PRRSV). Using this technique, the distribution and quantitation of PRRSV in naturally infected pigs were investigated. A standard curve (y = 4.94x + 0.18 ; R^2 = 0.98) for viral quantitation was established using target sequence and internal conrol plasmids constructed from a Taiwanese PRRSV isolate. Examination of the sera sequentially collected from piglets at various ages showed that the isolation rates of PRRSV reached the highest level of 100% in pigs at the age of 9 wks, which coincides with the lowest level of passive maternal antibodies. At this age, the mean concentration of PRRSV in the sera was 10^4.1 copies/mL. For pigs naturally infected with PRRSV, the lungs had the highest viral load with a range between 10^5.7 and 10^8.9 copies/g. The concentrations of the PRRSV in Iymphoid organs and sera were greater than 10^5 copies/g (mL). Nasal discharge and saliva had mean concentrations of 10^5.8 and 10^4.8 copies/mL, respectively. A range between 10^2.5 and 10^3.5 copies/mL of PRRSV was detected for the alimentary tract and urine. Results indicate that the loss of maternal antibodies increases the RRSVinfected populations in the pig herd. The major target organs for PRRSV are the lungs and Iympoid tissues. Nasal discharge and saliva are the main sources for the transmission of PRRSV.
