Comparison on Phagocytic Activity of Rainbow Trout (Oncorhynchus mykiss) Phagocytes Against Aquatic Bacterial Pathogens

The phagocytes in salmonid fish are believed to play the major role on their nonspecific immunity against most aquatic pathogens and irritants. In this trial, flow cytometric analysis was applied to detect and compare the phagocytic activity of rainbow trout (Oncorhynchus mykiss) phagocytes isolated from blood, head kidney and spleen, respectively. In general, the blood phagocytes showed higher capability of phagocytosis than spleen and head kidney phagocytes in engulfing Vibrio parahemolyticus, Vibrio anguillarum and Aeromonas hydrophilia. The results also demonstrated that bacteria opsonized with or without normal fish serum did not manipulate phagocytes to uptake and eradicate bacteria after 120 minutes incubation. However, the isolated phagocytes could enhance their phagocytic activity if increasing cultivation time from 2 to 24 hours. In addition, all phagocytes engulfed much more bacterial pathogen that could secrete hemolysin or their extracellular products (ECPs) could induce significant cytotoxicity on phagocytes.
