以鴿高病原性新城病病毒(NDV)株抗原製成不活化疫苗強化幼鴿對NDV之免疫保護力

選對台灣10個鴿群之164隻賽鴿血清可檢出較高血球抑制凝集反應(hemagglutination-inhibition; HI)抗體力價幾何平均值(geometric mean titer; GMT) 和變異系數(coefficient variation; CV)值較高的4株第VII型WNDV分別口服(1000 MLD)感染1組四週齡低NDV-HI抗體幼鴿,發現TW/99-V156B、TW/99-V157、TW/99V158和TW/84-486四株NDV分別可造成100%、60%、20%和0%死亡率與其在臨床檢出的NDV-HI抗體力價GMT和CV值高低頗有正相關聯性。以上述4株WNDV病毒液混合製成之NDV不活化油質與鋁膠型疫苗,併同市售6種鴿用NDV不活化疫苗分別免疫8組,每組6隻之四週齡幼鴿,於免疫25日後再以該4株NDV混合液口服(1000 MLD)攻擊,結果該NDV不活化疫苗對幼鴿之免疫保護力顯較其他市售鴿用NDV不活化疫苗為高。選用在臨床檢測鴿血清具高抗體之高病原性新分離WNDV株製成NDV不活化疫苗可有效強化幼鴿對NDV之免疫保護力。

Four isolates of genotype VII of velogenic viscerotropic Newcastle disease virus (WNDV) were selected on the basis of high geometric mean titer (GMT) and coefficient of variation(CV) values in sera from 10 flocks of racing pigeons; these isolates were used to inoculate 20 four-week-old pigeons (4 groups, 5 birds per group) orally with low NDV-HI titers. The mortality caused by the TW/99-V156B, TW/99-V157, TW/99-V158 and TW/84-486 isolates of WNDV was 100%, 60%, 20%, and 0%, respectively. The virulence of each isolate correlated positively with the GMT level and CV value of each isolate in assays with the above racing pigeon sera. Eight groups of 4-week-old pigeons (6 birds per group) with low NDV-HI antibody levels were immunized with either oil-emulsion or gel-type inactivated vaccines prepared with the above 4 isolates and or with one of 6 commercial inactivated NDV vaccines, then challenged orally (1000 LD50) with the mixture of the 4 homologous isolates at 25 days post vaccination. The inactivated vaccine prepared from the 4 WNDV isolates with high existing serum antibody and high virulence prominently provided higher protection levels than the commercial vaccines, and thus higher protection to pigeons.
