亞洲猴腦鹿角蕨分生苗量化繁殖途徑之研究

The study of in vitro mass propagation for mericlones of Platycerium ridleyi Christ

摘要


鹿角蕨(Platycerium)為近年來極受歡迎的熱帶觀賞蕨類植物,然而相關的組培研究有限。本研究將亞猴鹿角蕨(Platycerium ridleyi Christ)無菌孢子苗繼代於4種MS無機鹽類濃度(MS、1/2MS、1/4MS、1/8MS)之基本培養基,觀察其生長情形,結果顯示4種處理間有顯著差異,1/2MS處理組中增殖率及新葉數表現較佳且植株生長勢較好,建議以1/2MS無機鹽類為基本培養基。接續使用NAA(0、1、2 mg L^(-1))、BAP(0、1、2 mg L^(-1))、TDZ(0、0.5、1 mg L^(-1))及活性碳(0、0.1、1%),應用正交表設計出9種試驗組合,結果顯示幼孢子體在1/2MS基本培養基添加1 mg L^(-1)NAA、2 mg L^(-1)BAP及0.1%AC的條件下,分生苗增殖率可達100%,且生長較為快速、葉片發育良好,建議以此為鹿角蕨量化繁殖途徑之基礎。

Staghorn ferns (Platycerium Desv.) are very popular ornamental ferns in the recent years. In this study, in vitro sporophytes of Platycerium ridleyi Christ were used as plant materials and cultured on four basal medium containing different MS inorganic salts (MS, 1/2 MS, 1/4 MS, 1/8 MS). After culture for three months, 1/2 MS inorganic salt was screened as the optimal basal medium for plant growth and development of P. ridleyi. We also applied orthogonal table to design 9 combinations with NAA (0, 1, 2 mg L^(-1))、BAP (0, 1, 2 mg^(-1))、TDZ (0, 0.5, 1 mg L^(-1)) and activated charcoal (0, 0.1, 1%) for evaluating the effects of plant growth regulators and activated charcoal on sporophyte multiplication of P. ridleyi. We suggested that the combination of 1 mg L^(-1) NAA, 2 mg L^(-1) BAP and 0.1% AC is suitable for sporophyte multiplication with multiplication rate 100% and mericlones grew well. Tissue culture technology can be used to mass propagation of rare staghorn ferns with high value. In addition to save time and space, the application of plant tissue culture also cost down the input of labor with the advantages in mass propagation of rare staghorn ferns.
