山漆雙黃酮與紫外線暴露對肝癌細胞細胞週期之影響

Effects of Amentoflavone and Ultraviolet Exposure on Cell Cycle in Liver Cancer Cell

摘要


山漆雙黃酮(Amentoflavone, AF)乃來自裸子植物山漆果核萃取後之成分,具有抗病毒、抗肝炎及抗愛滋病毒等作用,紫外線(UV)是過氧化物物種主要的誘發劑及細胞凋亡的誘發因子,本實驗研究目的乃在釐清山漆雙黃酮與紫外線對於肝癌細胞合併處理下所誘發之細胞凋亡與細胞週期改變之影響。肝癌細胞(Hep G2 cell)對於山漆雙黃酮的敏感度不佳,以250μM山漆雙黃酮濃度下以流式細胞儀(flow cytometry)分析其存活率都高於80%以上。依此劑量下只輕微增加細胞週期中G2/M休止的現象,山漆雙黃酮與紫外線合併處理下明顯增加細胞週期中G2/M休止的現象,顯示可能伴隨cyclin B量的下降媒介而延生,硫胱氨酸蛋白酶-3(Caspase-3)和poly (ADP-ribose) polymerase (PARP)的表現也同樣產生下降的表現,其他週期與控制組並無明顯的差異,但凋亡誘發因子(Apoptotic inducing factor, AIF)的表現卻明顯增加,其合併死亡率的增加與硫胱氨酸蛋白酶-3可能無關,AIF的表現可能比較重要。

Amentofalvone (AF) was extracted from Rhus succedanea of gymnosperm. This component has many functions, including anti-HIV, anti-virus and anti-hepatitis. Ultraviolet B (UVB) is a ROS-induced agent and apoptosis induced agent. This study is to evaluate the combined effects of amentoflavone and UV on cell cycle and apoptosis in liver cancer cells. Hep G2 cell is not sensitive on amentoflavone treatment. Survival rates of 250μM AF treatment showed by flow cytometry kept over 80%. Amentoflavone or UV treatment itself, which is assayed by flow cytometry, got only a little enhancement of G2/M arrest. Moreover, G2/M arrest of the combined treatment with UV and AF was mediated by cyclin B decreases apparently. Caspase-3 and PARP expression were decreased. AIF expression enhanced slightly after combined treatment of amentoflavone and UV. Cell cycle assay was not increased more than the control group. It showed that combined group enhances cell death but not mediated through caspase-3 pathway of apoptosis. AIF expression may be important than caspase-3 pathway.
