不同年齡層注意力認知功能連結性變化:靜息態功能性磁振造影研究

Changes in Attentional Cognitive Function Connectivity in Different Age Groups: A Resting-state fMRI Study

摘要


當健康受試者在接受功能性磁振造影掃描時,我們可以經由分析休息狀態時的注意力功能連結性來觀察其認知功能的表現,透過此觀察,可以了解到在不同年齡下認知功能的表現。本研究的目的是要探討注意力功能在正常退化的過程中是否會隨著年齡的增長而影響到認知功能的表現。本研究總共招募了238名年齡介於5-80歲之間的健康受試者,所有受試者按15歲的年齡組距分為5個年齡層。每位受試者須接受時間六分鐘靜息態的功能性磁振造影掃描。選取雙側intra-parietal sulcus位置當作參考點,並以該點的時間反應曲線與全腦像素進行功能連結性計算,產生腦功能連結性腦圖。之後,以二因子變異數分析針對五個年齡層以及左右兩個功能網路來檢驗認知退化的變化情形。由我們的實驗結果可以推論出正常注意力退化確實會因為年齡的增長而影響,受到影響的腦區主要位在右側大腦的頂葉-枕葉交接區域,這個腦區跟注意力的切換有關。此外,我們的研究也觀察到,左右大腦認知功能的分工明顯發生在21-35歲的年齡層,而這個現象在51-65歲年齡層後漸趨平緩。本研究未來可提供大腦正常認知功能退化的模板供臨時病態性退化疾病診斷與治療的參考。

When healthy subjects were underwent a functional magnetic resonance scan, we can observe cognitive changes by analyzing changes in attention function during rest. The purpose of this study was to investigate whether the functional alteration of attentional cognition in the processing of normal aging. A total of 238 healthy subjects from 5 years old to 80 years old was enrolled in this study. All subjects were divided into 5 age groups by the interval of 15 years. Each subject at resting state was instructed to receive a six-minute functional magnetic resonance imaging scan. Afterward, a seed-based method was used to calculate the functional connectivity of attentional network for each subject based on two reference seeds at bilateral intra-parietal sulcus. As to statistical examination, a two-factor variance analysis method was performed to examine the effect of aging-associated factors according to five age levels and two functional networks. From our experimental results, it was inferred that the attentional function of healthy subjects is changed along with aging processing and more than that is affected by the distribution of brain function between hemispheres. The affected regions are located at the parietal-occipital junction of the right brain whose function is related to the switching of attention. Our result emphasized that the mechanism of diseased-caused cognitive impairment is available through the observation of normal aging processing.
