RAPD分子標誌應用於臺灣軟骨花蒜種原歧異度分析

RAPD Analysis on the Genetic Diversity among Germplasm of Taiwan Soft-stem Garlic (Allium sativum)

摘要


本研究利用RAPD(random amplified polymorphic DNA)分子標誌來進行臺灣本土軟骨花蒜地方種(宜蘭花蒜、安南區花蒜、花蒜選、花蒜、花蒜早熟種)之種原歧異度分析。試驗結果顯示自15組逢機引子(random primers)中篩選出4組具多型性之標誌引子(OPAB4、OPC5、OPC7與OPC13),共產生93組DNA條帶,其中多型性DNA條帶有52組,具有55.9%多型性。於群聚分析上,遺傳距離介於52~81%,同時亦可將供試地方種明顯區分為A、B、C三群。藉由RAPD分析之結果顯示,此五種花蒜地方種並非相同品種且存在有相當之歧異度。

Random amplified polymorphic DNA (RAPD) analysis was employed to evaluate genetic diversity among five Taiwan endemic soft-stem garlic varieties Hua-suan (Allium sativum), namely, Yuan Hua-suan, Annan area Hua-suan, Hua-suan Selected, Hua-suan, Hua-suan early maturity. Four (OPAB4, OPC5, OPC7 and OPC13) out of the fifteen synthetic random primers were selected to identify polymorphism in amplification products. Of a total 93 bands, 52 were polymorphic, with 55.9% polymorphism among the five garlic varieties. Custer analysis revealed that the range of genetic diversity was between 52-81%, which could divide the five garlic varieties into three groups (A, B and C groups). According to the results of RAPD analysis, these five Hua-suan garlic varieties may not be the same species and existing genetic variation.
