生長調節物質對於大蒜培殖體生長之影響

Effect of Growth Regulators on the Growth of Garlic Explants

摘要


本研究以TA350及TA613二個大蒜種原的培殖體為材料,將其培養在僅含30、80及100 gL^(-1)蔗糖的MS起始培養基(initial MS medium)中。經過3個月後,再分別移植至蔗糖濃度為30、80、100 gL^(-1)及分別含1、10及50 ppm SADH (succinic acid-2,2-dimethyl hydrazide)或MH (maleic acid hydrazide)或CCC (2-chloroethyl-trimethylammonium chloride)的MS培養基中,以期能降低植株生長速率且不影響存活率的條件下,篩檢出最適當延長繼代培養的條件,以達到大蒜中期種原保存的目標。由研究發現,TA350及TA613大蒜培殖體培養在30 gL^(-1)蔗糖配合50 ppm SADH或1ppm MH或50 ppm CCC之處理組,植株生長緩慢且具有較高的成活率。

The study was aimed to determine the best combination of sucrose concentration and growth regulator of MS medium for reducing the growth and giving the longest cycle of time per subculture. Two accessions of garlic explants grown in vitro were used in the experiment, namely, TA350 and TA613. The explants were cultured in the initial MS medium which contains 30, 80 and 100 gL^(-1) sucrose for 3 months, then transplanted to the 3 concentrations (30, 80, and 100 gL^(-1)) of sucrose together with different concentrations (1, 10, and 50 ppm) of SADH (succinic acid-2,2-dimethyl hydrazide), MH (maleic acid hydrazide) and CCC (2-chloroethyl-trimethylammonium chloride), to determine the best combination for the germplasm conservation of garlic in vitro. The results indicated that the plantlets grown in 30 gL^(-1) sucrose together with 50 ppm SADH or 1 ppm MH or 50 ppm CCC showed reduced growth rate in vitro and high survival rate after transplantation.
