松茸接種於濕地松可行性之研究

摘要


松茸(Tricholoma matsutake Singer)是一種高經濟價值的食用及藥用真菌,由於能與松茸形成共生狀態的宿主有限,且松茸對於生育地環境因子要求嚴格,因此人工栽培松茸不易。本研究分別以濕地松(Pinus elliottii Engelm.)組織培養苗及無菌實生苗接種松茸菌絲,透過接種苗及未接種對照組苗木之間新生一次針狀葉(primary needle)數目差異之比較及生物量(苗木乾重)的改變,以及藉由顯微鏡輔助觀察接種苗形成菌根(mycorrhiza)之特殊形態結構,探究松茸接種於濕地松之可行性。試驗結果顯示,接種苗因為有菌根存在,其生長顯著優於未接種苗。感染松茸的濕地松幼根表面被覆松茸菌絲,其中具有末端膨脹之氣生菌絲,但尚未觀察到菌毯(mentle)的形成。感染部位呈暗褐色,根尖呈分支狀,皮層細胞內觀察到類似澱粉粒之小顆粒等外生菌根特有共生構造。因此,本研究證實濕地松亦能被松茸感染形成外生菌根。

Tricholoma matsutake Singer is an edible pine mushroom of high economic value and with medicinal properties. However, it is difficult to be cultivated commercially due to its limited symbiosis with host species and specific requirements of habitat. This study investigated the feasibility of Pinus elliottii Engelm. (tissue culture propagated plantlets and in vitro grown seedlings) inoculated by T. matsutake through the measurement of growth progress (i.e., number of primary needles and dry weight of plants) and the observation of mycorrhizal morphology on the inoculated plants by microscopes. Results showed that mycorrhizal morphology on the inoculated plants was evident and vegetative growth of inoculated plants was significantly greater than those of non-inoculated controls. The roots of infected pine were dark brown in color, bi-branched at tip end, and surface colonized by white external mycelium with occasionally observed aerial hyphae which can be identified by its terminal swelling. However, the mentle of the ectomycorrhiza was not observed. The starch grain-like particles were accumulated within the cortex cells of the infected roots. All of the characteristics observed were specific morphological structures or discernable features of symbiosis of ectomycorrhiza with host plants. Therefore, this study demonstrates that T matsutake can form an ectomycorrhizal structure on Pinus elliottii.
