米穀預拌粉與米食製品中米含量檢驗技術之開發

Methods Development for Determination of Rice Content in Pre-mixed Rice Flour and Its Related Processed Food

摘要


有鑑於國內目前正積極開發多元化米製品的生產技術,透過以米穀粉取代麵食製品中部分的麵粉,達到減少小麥進口,提昇國產米食消費量與提高糧食自給率的目標。同時為提升民眾對餐飲食米來源之信心,建立臺灣米優質形象,在提高米食消費之同時,極需建立配套的米穀預拌粉和米食製品中米含量檢驗技術,以利米標章之推廣及米食市場之管理與輔導。本研究利用生物資訊,以文獻搜尋及蛋白質資料庫比對方式,挑出水稻特有之專一性種子蛋白,合成短胜肽製備多源及單株抗體。針對水稻特有榖蛋白,實驗結果顯示其抗體專一性靈敏度佳,不會與大麥、小麥、玉米等穀物蛋白有交叉反應。據此我們開發出膠體金免疫法(colloidal gold-based immunochromatographic assay(CGIA))快篩試劑組及酵素結合免疫吸附分析法(sandwich ELISA;enzyme-linked immunosorbent assay)之定量檢測方法。可有效用於檢測市售之米穀預拌粉與不同米食製品,如爆米香、米花糖、米果、仙貝、米粉、米麵條、米麵包及米蛋糕等。此些技術有助於國內市場食米品質管制機制之建立,提供廠商原物料驗收、政府撥售加工用米流向控管及未來推動國產米加工品認證標章品質管理之依據。

Currently the production technology of diversified rice products has been actively developed. To reduce wheat imports, increase the consumption of domestic rice and the self-sufficiency rate of food, part of the flour in the pasta products is tended to be replaced by rice flour. At the same time, while promoting the consumption of rice, in order to enhance the confidence of consumers in the dietary value of rice and to establish a high-quality image of Taiwan rice, the government needs to establish an inspection technology for measurement of rice content in rice grain premixed powder and rice food products. In this study, bio-informatics approach was used to select specific seed proteins which are unique to rice through literature search and protein database comparison, then short synthetic peptides were prepared for polyclonal and monoclonal antibodies generation. The experimental results showed that the monoclonal antibody which specifically recognized rice glutelin protein is sensitive enough and would not cross-react with proteins from barley, wheat and corn grains. Based on this, we developed a colloidal gold-based immunochromatographic strip assay (CGIA) for rapid screening and a sandwich ELISA (enzyme-linked immunosorbent assay) for quantitative measurement of rice content. These two methods can be effectively used to detect the commercially available rice premix flour powders and different rice food products, such as popcorn, rice candy, rice cracker, senbei, rice noodles, rice bread and rice cake. These technologies contribute to the establishment of a quality control mechanism for rice in the domestic market, providing the basis for the acceptance of raw materials checkout by the manufacturers, the control of the flow of processed rice, the management of the quality of domestically processed rice products and to promote certified quality monitoring in the future.
