中藥藥材六味地黃丸、菊杞地黃丸與養血壯筋丸經鈷-60輻射滅菌後之致突變試驗研究

摘要


本文敘述利用基因轉殖後的細菌株(沙門氏桿菌),在遭受新的外來物質刺激下,可用於偵測生物性混合物之檢體安氏-沙門氏桿菌/微粒體致突變性試驗法。中藥丸劑經輻射照射滅菌後,是否會造成中藥性質改變也可藉由此安氏試驗法加以鑑定。本實驗以中藥丸-六味地黃丸、菊杞地黃丸與養血壯筋丸等三種丸劑分別經過。kGy、5kGy、10kGy、19kGy、25kGy、30kGy的60Co加馬照射後,利用基因轉殖後之沙門氏桿菌屬TA98與TA100兩種菌株進行偵測。TA98用來偵測引起結構移位(frame-shift)致突變物,TA100是偵測因起G-C鹼基對更換(base-pair substitution)。經實驗證實,此三類中藥藥丸-六味地黃丸、菊杞地黃丸與養血壯筋丸,在經過鈷-六十加馬射線照射後0~30kGy的照射劑量下均不致造成致突變性。

This paper reported the use of transformed salmonella (TA100 and TA98) in the mutagenesis test for the radiated Chinese medicines. In this experiment the TA100 strain is used to detect the base-pair substitution mutagenesis, and the TA98 strain is used to detect the frame-shift mutagenesis. The Chinese medicals were irradiated for 0kGy, 5kGy, 10kGy, 18kGy, 25kGy and 30kGy. Each medical was then mixed with the transformed salmonella (TA100 and TA98) and transfer into the low glucose media plate. After incubated at 37℃ for 48~72 hours, the Ames standard protocol is used to analysis the mutagenesis ratio. To colony number growth on the plate calculated to make the score of the test In Ames test we could not found any different mutagenesis between the controls and radiated Chinese medical. So it was concluded that the radiation won't induce any mutagenesis to the Chinese Medical.
