The present study demonstrates that milkfish retorted skin gelatin hydrolysates (RSGH) possess antioxidant activities including DPPH scavenging, reducing power, lipid peroxidation inhibition, and protective effect of DNA oxidative damage. The antioxidant activities of RSGH derived from different levels of thermal hydrolysis (from 0.5 to 3 h) showed skin gelatin retorted for 3 h (RSGH-3h) had the highest antioxidant activity. Besides, the main molecular weight ranges of the RSGH-0.5h RSGH-1h, RSGH-2h, and RSGH-3h were>6511 (62.48%), 686~12500 (87.49), 189~6511 (81.39%), and 189~6511 Da (84.44%), respectively. After sequential ultrafiltration with various MWCO ranges, 1k-RSGH-3h ultrafiltrated from RSGH-3h was the most strong antioxidant activity among 1k-RSGH-3h (MWCO<1kDa), 5-1k-RSGH-3h (MWCO: 1~5 kDa), 10-5k-RSGH-3h (MWCO: 5~10 kDa), and 10k-RSGH-3h (MWCO>10 kDa). It indicated that oligopeptides in RSGHs play an important role in their antioxidant activities. Moreover, milkfish 1k-RSGH-3h shows great potential as a natural antioxidant.
