探討樟芝萃取物調控人類大腸癌細胞凋亡之訊息路徑研究

The Signal Transduction Pathway of Apoptosis by Antrodia Camphorate Extracts in Human Colon Cancer Cell

摘要


目的:中草藥萃取物中以樟芝多醣體萃取物之生物效果最佳,研究指出樟芝的生物效果以抗氧化(antioxidant)、解毒(antidote)、抗癌(anticancer)、止癢(antiitching)、保護肝臟及免疫調節(immunomodulatory)為主;然而對大腸癌細胞的抑制效果則未曾有系統的研究,臨床上大腸癌的發生率近年來有顯著上升的趨勢,因此如何預防與治療大腸癌已成為重要的課題。方法:本計劃以SW620 cell line(colon adenocarcinoma cell line;人類結腸癌細胞株)進行體外培養,探討樟芝萃取物對於大腸癌細胞凋亡(apoptosis)訊息路徑的影響,我們以流式細胞儀分析癌細胞之細胞週期、細胞凋亡以及使用西方墨點法分析細胞凋亡相關訊息路徑變化情形與下游蛋白質的表現,以進一步探討樟芝萃取物抑制大腸癌細胞之機轉。結果:研究結果顯示,細胞再加入不同濃度之樟芝萃取物後,經flow cytometry分析後發現樟芝萃取物明顯發細胞凋亡反應,顯示樟芝萃取物確實具有促使癌細胞細胞凋亡之能力,然後經西方墨點法分析後發現,樟芝萃取物亦明顯誘發細胞凋亡蛋白caspase3/7/9與PARP(Poly (ADP-ribose) polymerase)等,研究結果顯示樟芝萃取物藉由誘發細胞凋亡蛋白之作用機轉來達成抑制癌細胞之效果。結論:因此本研究成功證實樟芝萃取物具有抗大腸癌細胞之能力與其可能之抗癌作用機轉,本研究不止促進抗癌細胞相關機制的瞭解,亦可開發另一種抗癌之輔助癌法。

Objective: Chinese Herbs extracts were proved be capable of free radicals cleavage, anti-carcinogenicity, enhancement on human immunity. Antrodia camphorate (Chinese name, niu-chang-chih or chang-chih) is an exclusive fungus parasitic on the inner cavity of the endemic species Cinnamomum kanehirai Hayata and an important traditional Chinese medicinal fungus (Basidiomycetes) for the treatment of human diseases such as food and drug intoxication, diarrhea, abdominal pain, hypertension, itchy skin and liver cancer. The anti-cancer by Antrodia camphorate extracts against colon cancer were never been systemic evaluated. The purpose of this study is to verify the inhibitory effect on colon carcinogenicity by Antrodia camphorate extracts.Methods: The evaluation methods including modulation of signal transduction of apoptosis, shift of cell cycle by FACscan flow cytometry, and caspase, parp analysis by western blots. The mechanisms of anti-carcinogenesis, evaluation models and new function-potential screening standard process will be established on Antrodia camphorate extracts in this study.Results: The results indicated the amounts of caspase3/7/9, parp and the apoptosis effects were dose-related induced. The result of present study can provide the experimental evidence that inhibitory effect on anti-carcinogenicity of colorectal adenocarcinoma by Antrodia camphorate extracts.Conclusions: The potential benefits of anti-cancer drugs production value will be promoted and enhances the international competitiveness largely and will prove to be helpful for understanding the anti-carcinogenesis mechanics by Antrodia camphorate extracts and provide novel insights in controlling colon cancer in our population.
