應用ITS序列鑑定天南星藥材及其製劑

Identification of Arisaematis rhizoma and Its Preparations by Nested PCR-amplified ITS Region Sequencing Method

摘要


天南星藥材基原為天南星科植物中,異葉天南星(Arisaema heterophyllum)、天南星(Arisaema erubescens)或東北天南星(Arisaema amurense)之乾燥塊莖。由於本身具有毒性,為避免藥材汙染或誤用造成之健康危害,本研究選擇以internal transcribed spacer (ITS)序列作為基因標記,設計天南星專一性引子組,並利用Nested PCR繼以DNA定序法進行基原鑑別。測試所收集之天南星及其誤用品藥材(含7個品種總計36件藥材)以及4件複方製劑檢體,結果證明此方法不但能有效地應用於未經炮製或各種炮製方法處理後的藥材,亦可應用於複方製劑之檢驗。

Arisaematis rhizoma is the dried tuber of three Araceae species, namely Arisaema erubescens (Wall.) Schott, Arisaema heterophyllum Blume and Arisaema amurense Maxim. To avoid misusing this toxic material, a nested PCR-DNA sequencing method for the identification of raw materials, refining materials or preparations of arisaematis rhizome was developed based on the sequence of internal transcribed spacer (ITS) region. In this study we analyzed 36 herbal samples of 7 species and 4 traditional Chinese Medicine (TCM) preparations, and the results showed this method provides effective and accurate identification of arisaematis rhizome.
