胡瓜子葉核糖核酸之代謝

Ribonucleic Acids Metabolism during the Development of Cotyledons of Cucumis Sativus L.

摘要


以胡瓜(Cucumis satiuas L.)為材料,研究種子萌發初期及幼苗生長過程中子葉核糖核酸之代謝。在種子萌發的各階段中,將其子葉與胚的其他部份分離,並用酚液抽取其RNAs。將所抽取之RNAs以MAK(methylated albumin kieselguhr)圓柱色層分析法分析其各類RNA之成分。在種子萌芽期中各類RNA之含量有顯著改變。以放射磷-32作追踪劑之實驗中,rRNA(ribosomal RNA)和sRNA(soluble RNA)之合成最為迅速。將胡瓜種子分別在光與暗中萌發時,其子葉RNAs以Acrylamide gel電泳法分析,結果顯示出葉綠體rRNA和細胞質rRNA之合成受光之刺激而增加,而低分子量(low molecular weight)RNA則不受影響。

Changes in ribonucleic acids (RNA) fractions of Cucumis sativus L. during germination were studied. At various stages of seed germination, cotyledons and other parts of embryos were separated and RNAs were extracted by a phenol-method. Total RNAs were fractionated on the methylated albumin keselguhr (MAK) columns. As seed germination, proportions of RNAs in embryos changed significantly. In cotyledons, synthesis of rRNA occured in the early stages, 6 to 24 hr after the onset of imbibition. 32P was also incorporated in sRNA continuously at rather a moderate rate. RNAs isolated from light-grown and etiolated cotyledons were tractionated by polyacrylamide gel electrophoresis. The results show that ribosomal and low molecular weight RNAs of chloroplast are different from that of cytoplasm. If 32P is used as precursor of RNA, then light stimulated the Incorporation into chloroplast and cytoplasmic RNAs but no significant change in low molecular weight RNAs.
