臺灣產五種常見蚊蟲異構酶組型之研究-第一報 酯酶及乳酸去氫酶之組型

摘要


與日本腦炎傳佈有關,於實驗室中培育數年之五種臺灣產蚊蟲,三斑家蚊、鹹水家蚊、環紋家蚊、熱帶家蚊及白腹叢蚊,以聚丙稀醯胺膠體電泳法分析其第四齡幼蟲,蛹及成蟲期酯酶(esterase)及乳酸去氫酶(lactate dehydrogenase)之異構酶組型,發現該兩種酶之活性於第四齡幼蟲期最高,於蛹及成蟲期則減弱。酯酶異構酶之組型相當複雜,各蚊種均有其特異性;即使同一蚊種,其組型亦隨發育時期之不同而發生變化;三斑家蚊更具雌雄性別不同之組型。乳酸去氫酶之組型較單純,唯大異於哺乳類之此酶組型。該酶亦具種之特異性,然三斑家蚊、鹹水家蚊及環紋家蚊第四齡幼蟲期組型之相似性,顯示歸屬同一族(series)之蚊蟲,不但形態類似,在生化學上或許亦具相似性。

Esterase and lactate dehydrogenase (LDH) isozyme patterns were studied from five common species of laboratory colony culicine mosquitoes, may be involved in transmission of Japanese B encephalitis. By using 7.5% polyacrylamide gel electrophoresis, the zymograms proved to be species-specific, and changed during development. Sex dimorphism was shown to occur in the esterase zymogram of one species, Culex tritaeniorhynchus summorosus. The enzyme activity was the highest at the fourth instar larval stage and decreased at pupal and adult stages. Zymograms or lactate dehydrogenase of 3 species, Culex tritaeniorhynchus, Culex sitiens, Culex annulus, were also shown to be very similar at 4th instar larval stage. This similarity of LDH isozyme patterns may provide biochemical evidence to support the classification system which classifies them into the Culex sitiens series in Culicinae (Diptera: Culicidae).
