二種單胞固氮綠藻之形態與生理特性的比較

Characterization of N2-Fixing Unicellular Cyanobacteria Synechococcus RF-1 and Gloeocapsa RF-6

摘要


聚球藻RF-1(SynechococcusRF-1)與黏球藻RF-6(Gloeocapsa RF-6)為分離自臺灣南部水田的單胞固氮藍綠藻,是革蘭氏陰性,採二分法繁殖的微生物。兩者均具有微弱的觸酶活性,但不具過氧化酶和酪蛋白水解酶的活性。眾球藻RF-1的顏色為墨綠色,細胞呈橢圓形,長軸約4μm,短軸約3μm,細胞外沒有鞘的構造,但具有微弱的澱粉酶活性。黏球藻RF-6的顏色為土綠色,細胞呈圓球形,直徑約5μm,具有發達的鞘,但測不到澱粉酶的活性。培養於含氮源之培養基(BG-11)時,聚球藻的觸酶活性以及藻紅素與藻藍素的含量,均較無氮源(BGo-11)培養時提高很多。兩者對重金屬離子的敏感性相似,其中以對汞的敏感性最高,致死濃度為0.01ppm,其次依序為銅、銀、鉛;對於砷則有很高的耐性,致死濃度高達10^5 ppm。

Synechococcus RF-1 and Gloeocapsa RF-6 were unicellular N2-fixing cyanobacteria purified from the rice field of southern part of Taiwan. They were Gram negative and divided by binary fission. They had weak catalase activities, similar sensitvities to heavy methals, but no casein hydroiyase and peroxidase activities. Synechococcus RF-1 had dark green color, elliptical shape, and without sheath. It contained weak amylase activity. Gloeocapsa RF-6 had yellow green color, spherical shape, and sheath-containing. But Gloeocapsa RF-6 did not have amylase activity. The presence of nitrogen source in medium elevated the catalase activity, and the content of phycoerythrin and phycocyanin. Syneclrococcus RF-1 and Gloeocapm RF-6 were very sensitive to Hg^2+, Cu^2+, Ag^1+, and pb^2+, but rather tolerant to As^5+.
