以分子標記鑑定食用紅甘蔗品系初探

An Attempt to Identify Edible Sugarcane (Saccharum officinarum L.) of Different Local Lines with Molecular Markers

在不同地區栽培品系基因差異性之分析上,本研究選殖食用紅甘蔗核糖體基因內轉錄間隔區(Internal transcribed spacer,ITS),再以限制脢(restriction endonuclease)加以切割,在16個品系中,初步獲得57個條帶,其中有13個條帶為專屬條帶,分別屬於二水、油車、關山及池上。其中以Hpa II可直接鑑定出關山及池上品系。顯示東部地區之栽培品系與西部之栽培品系有明顯差異,而多形性片段顯示不同地區栽培品系確有不同。

On the analysis of genetic difference of different local lines of edible sugarcane, the internal transcribed spacer (ITS) of ribosome DNA (rDNA) was cloned and cleavage with restriction endonucleases. Among the 16 lines tested, total 89 bands were obtained and in which 13 bands are unique to Guan-Shan (5) and Chi-Sang (8). With HpaII, local lines of Guan-Shan and Chi-Sang could be distinguished from the lines cultivated in western Taiwan, indicating that there are considerable differences exist between eastern and western populations. The polymorphic band patterns indicate that there are some differences among lines cultivated in different districts.
