大豆發酵液之免疫調節功效

The Immunomodulatory Effect of Soybean Fermented Fluid

摘要


大豆(Glycine max (L.) Merrill, 亦稱黃豆),研究證實它富含蛋白質、必需氨基酸、鈣、鐵、維生素(A 與B)以及植物油等。本研究評估大豆發酵液對抗原呈獻細胞(antigen-presenting cells;APCs)如樹突狀細胞(dendritic cell;DC)、巨噬細胞(macrophage)和B細胞與T細胞之間所產生的作用及其相關細胞激素的產生。應用BALB/c小鼠餵食三種大豆發酵液劑量(0.288、0.864、1.440 g/kg)達二、四、六週,於服用後不同時間點(24、72、120小時)進行犧牲取其脾臟細胞進行分析。研究結果顯示,大豆發酵液能明顯提升抗原呈獻細胞數量以及其上表面分子CD8、CD40、MHC clsassⅡ(I-A/I-E)等之表現增加,顯示餵食後能夠促使抗原呈獻細胞之成熟與活化,且可見與T細胞上之共同刺激分子相互作用。篩選對活化免疫細胞表現之結果最佳的高劑量(1.44 g/kg)大豆發酵液,應用Real-time PCR分析相關細胞激素mRNA之表現,研究發現能提升增強BALB/c鼠體內T_H1細胞激素INF-γ、IL-12及巨噬細胞分泌TNF-α之表現,相對的抑制T_H2細胞所分泌細胞激素IL-4表現。研究結果顯示大豆發酵液促使免疫走向T_H1細胞免疫反應,能有效提升免疫力之功能。

Soy [Glycine max (L.) Merrill, also known as Soybean], is rich in protein, amino acids, calcium, iron, vitamin A & B as well as vegetable oil. This research investigates the immunomodulatory activities and the cytokines produced by soybean fermented fluid to antigen-presenting cells (APCs) such as dendritic cells (DC), macrophage and B cell which interacted with T cells. Feeding BALB/c mice with three different concentrations of soybean fermented fluid (0.288, 0.864 and 1.440 g/kg) over a 2, 4, or 6-week period while taking spleen cells at 24, 72, or 120 hours after feeding for performing experiments. The results showed that soybean fermented fluid will increase the amounts and the expression of surface molecule CD8 、CD40、MHC class II (I-A/IE) on APCs, indicating the maturation and activation of APCs and the interaction with T cell's co-stimulatory molecules after feeding. The highest concentration of 1.44 g/kg obtained the best activation for immune cells as performed with further real-time PC R analysis of cytokine mRNA gene expression. The results showed that soybean fermented fluid can increase the expressions of TH1 cytokine INF- IL-12 and macrophage secreting TNF- in BALB/c mice with relative suppression of TH2 cytokine IL-4. These data strongly suggest that soybean fermented fluid is able to increase the host immunity by inducing TH1 immune responses.
