利用線上衍生化系統結合液相層析儀檢測食品中α-胺基丁酸的含量

Detection and Quantification of γ-Aminobutyric Acid in Health Food by Using Online Derivatization System Coupled with HPLC

摘要


γ-胺基丁酸(γ-aminobutyric acid, GABA)具有在改善失眠、抑鬱和更年期綜合症等方面有顯著效果,利用微生物發酵生產GABA,具有成本低、含量高及可安全且可用於食品的優點,目前常將GABA作為天然功能性食品添加於保健食品中。由於α-胺基丁酸(α-Aminobutyric acid, AABA)、β-胺基丁酸(β-Aminobutyric acid, BABA)與GABA為同分異構物(isomers),但具有不同的功能,因此有必要將三者加以區分。本研究利用線上衍生化系統(online derivatization system)搭配高效液相層析儀紫外光檢測器(high performance liquid chromatography/diode array detector, HPLC/DAD)鑑定AABA、BABA 及GABA。線上衍生化系統衍生化流程為將衍生化試劑放置於儀器內的自動化取樣系統(multisampler)中,接著以軟體控制機械手臂將樣品與衍生化試劑混合。此方法能減少以往傳統衍生化(traditional derivatization system)所要的時間及人為操作誤差。在GABA結果顯示,當濃度在4~84 ppm的校正曲線(standard curve)呈線性,其相關係數(correlation coefficient, r)為0.9994。於樣品分別添加4 ppm、8 ppm與12 ppm的GABA,其等之回收率(Recovery rate)皆在110%內,重覆性(repeatability)及中間精密度(intermediate precision)之變異係數(coefficient of variation, CV)分別小於2%與3%,這些效能皆符合食品化學檢驗方法之確效規範。由於檢測市售保健產品中GABA含量與標示量一致。上述的發現證明此分析方法可應用於檢測保健食品中GABA的含量。

Gama-aminobutyric acid (GABA) has beneficial effects in the treatment of insomnia, depression, and menopausal syndrome. Industrial GABA is usually produced by microbial fermentation because of low cost, high yield, and high quality. GABA is present in some natural foods and is a common additive in health foods. Since α-aminobutyric acid (AABA), β-aminobutyric acid (BABA), and GABA are isomers with different functions, it is necessary to distinguish them. In this study, an online derivatization system coupled with high performance liquid chromatography/diode array detector (HPLC/DAD) was used identify AABA, BABA, and GABA. The derivatization reagents were placed in the multisampler of the instrument. Samples were then mixed with the reagents by a software controlled robotic arm to initiate the complicated derivatization and analysis processes. This automatic system greatly reduced the time required for traditional GABA derivatization and operation errors. In the concentration range from 4 to 84 ppm, the standard curve for GABA was linear and the regression equation was obtained with correlation coefficient of 0.9994. When samples were spiked with 4 ppm, 8 ppm, or 12 ppm of GABA, the recovery rates were all approximately 110%. The coefficient of variation (CV) of repeatability and intermediate precision were less than 2% and 3%, respectively. These efficacies meet the requirements for food safety testing. As the GABA contents detected in commercial health food products with this system were consistent with those of the indicated amount, we conclude that this method is effective for detection and quantification of GABA in health food products.
