綠膿桿菌在Mueller-Hinton Medium、Cetrimide Agar與CN Agar產Pyocyanin(綠膿素)與Fluorescein(螢光素)的能力

Production of Pyocyanin and Fluorescein by Pseudomonas aeruginosa in Different Brands of Mueller-Hinton, Cetrimide Media and CN agar

摘要


Cetrimide agar(溴化十六烷基三甲胺瓊脂)或cetrimide nalidixic acid(CN)agar係用於分離藥品、化妝品或礦泉水中之Pseudomonas aeruginosa(綠膿桿菌,銅綠假單胞菌),在Cetrimide agar與CN agar產生pyocyanin(綠膿素)與fluorescein(螢光素)能力為P. aeruginosa之主要特性。另外,Pseudomonas agar P與Pseudomonas agar F則用於分別確認pyocyanin與fluorescein的產生能力。吾等發現P. aeruginosa在臨床檢驗室常規操作紙錠瓊脂擴散藥敏試驗的Mueller-Hinton(M-H)medium亦能產生pyocyanin與fluorescein,但不同廠牌產生pyocyanin與fluorescein的能力及程度變化很大。本研究利用90株P. aeruginosa分別以等量接種不同廠牌(美國BD-BBL^(TM)、英國Oxoid與德國Merck)粉末所配製的M-H medium培養基,然後觀察其等產生pyocyanin與fluorescein的能力。結果發現以BD-BBL^(TM)的M-H medium產生pyocyanin與fluorescein的效果最佳,分別達到86.6%與82.2%的陽性率;Oxoid廠牌則次之,分別為75.5%與77.7%;Merck廠牌最差,分別為57.7%與44.4%。另外,CMP^(TM) CN agar產生pyocyanin 與fluorescein的能力(分別為96.6%與91.1%)明顯地優於兩種廠牌的cetrimide agar以及Pseudomonas A gar P與Pseudomonas Agar F。因此,選擇正確廠牌的M-H medium以及適當種類的CN agar與cetrimide agar將可更快速且準確地偵測/鑑定臨床檢體、藥品/化妝品或礦泉水中的P. aeruginosa。

Cetrimide (hexadecyltrimethylamine bromide) agar and cetrimide nalidixic acid (CN) agar are selective media for detection of P. aeruginosa in pharmaceuticals, cosmetics or drinking mineral water. The ability to produce pyocyanin and fluorescein on cetrimide agar and CN agar is a characteristic feature of P. aeruginosa, and Pseudomonas Agar P and Pseudomonas Agar F are used to confirm pyocyanin and fluorescein production, respectively. We have found that P. aeruginosa can also produce pyocyanin and fluorescein in Mueller-Hinton (M-H) medium, which is routinely used for antimicrobial susceptibility tests. However, pyocyanin and fluorescein production by P. aeruginosa in different brands of media varies greatly. In this study, we tested the ability of 90 strains of P. aeruginosa to produce pyocyanin and fluorescein in various media, including CMP^(TM) CN agar, CMP^(TM) cetrimide agar, BD-Difco^(TM) cetrimide agar, and M-H media from BD-BBL^(TM) (United States), Oxoid (United Kingdom), and Merck (Germany). Results showed that the M-H medium from Merck is not appropriate for detection of pyocyanin and fluorescein production by P. aeruginosa as the positive rates were relatively low. CMP^(TM) CN agar was found to be the best for pyocyanin and fluorescein production by P. aeruginosa, followed by two brands cetrimide agar. Pseudomonas agar P and Pseudomonas agar F are not good media for detection of pyocyanin and fluorescein, respectively. Therefore, select the right brand of M-H medium and the appropriate type of CN agar or cetrimide agar will enable faster and accurate detection/ identification of P. aeruginosa in clinical samples, pharmaceutics/cosmetics or mineral water.
