網狀紅血球為仍在發育的紅血球(red blood cells, RBC)。網狀紅血球的定量通常在採集血液樣本後3小時內進行,如果樣本延遲運送超過3小時的限制,則需要重新採檢。本研究調查在不影響網狀紅血球計數準確性的情況下儲存血液樣本的條件,共採集了39份血液樣本。每個樣本在採集後3小時內完成常規RBC與網狀紅血球計數,然後均分到兩個試管中。其中一支試管在室溫儲存,另一支在4°C儲存,然後分別在6和8小時的時間點使用Sysmex XN分析儀檢測樣本。結果顯示在收集血液樣本後3小時內與在室溫或4°C儲存6或8小時後所測定的網狀紅血球計數和網狀紅血球生成指數(reticulocyte production index, RPI)沒有顯著差異。因此,我們得出結論:血液樣本中的網狀紅血球在室溫或4°C下穩定至少8小時,如果在此期限內完成網狀紅血球計數,則結果有效。
Reticulocytes are red blood cells (RBC) that are still developing. Quantitation of reticulocytes is usually performed within 3 hours after a blood sample is obtained, and a retest is required if sample delivery is delayed beyond the 3-hour limit. In this study, we investigated the conditions in which blood samples can be stored without affecting the accuracy of reticulocyte counting. A total of 39 blood samples were collected. Each sample was equally divided into two tubes after routine RBC and reticulocyte counting was completed within 3 hours of sample collection. One of which was stored at room temperature, and the other was store at 4°C. The samples were then examined using the Sysmex XN analyzer at the time points of 6 and 8 hours. Results showed that there was no significant difference in reticulocyte counts and reticulocyte production index (RPI) of a sample determined within 3 hours after collection or after 6 or 8 hours of storage at room temperature or 4°C. Therefore, we conclude that retic ulocytes in a blood sample are stable at room temperature or 4°C for at least 8 hours and that results are valid if reticulocyte counting is completed within this time limit.
