從牛樟芝(Antrodia cinnamomea)菌絲體萃取的純化4-乙醯基蒽醌B(4-AAQB)具有抑制腦膜癌循環腫瘤細胞的潛力

Inhibitory Effect of Purified 4-Acetylantroquinonol B Form Antrodia cinnamomea Mycelium on Meningeal Carcinoma Cells

摘要


腦瘤的發病率為每10萬人中有4-5例,致死率高且預後差。本研究中,吾等從惡性腦膜瘤患者的血液中獲得循環腫瘤細胞,並且以牛樟芝(Antrodia cinnamomea)菌絲體之純化化合物:4-Acetylantroquinonol B(4-AAQB)測試其對這種細胞的抑制作用。結果顯示,4-AAQB在4μg/mL的濃度下對循環腦膜腫瘤細胞的生長有約50%的抑制作用,在20μg/mL的濃度下則有65%。結果亦顯示,4-AAQB和腦膜癌治療藥物Lomustine的併用並沒有協同作用。另外,還發現,4-AAQB可抑制LPS誘導的單核細胞產生一氧化氮。綜上所述,本研究結果指出,從牛樟芝菌絲體中純化的生物活性化合物4-AAQB具有抑制腦膜癌生長的潛力。

Brain tumors have an incidence of 4-5 cases per 100,000 people with a high fatality rate and poor prognosis. In this study, we obtained circulating tumor cells from patients with malignant meningioma and tested whether the purified compound, 4-Acetylantroquinonol B (4-AAQB), from Antrodia cinnamomea mycelium has inhibitory effect on these cells. Result showed that 4-AAQB inhibited the growth of circulating meningeal tumor cells by approximately 50% at the concentration of 4 μg/mL and 65 % at 20 μg/mL. Our results also showed that combined usage of 4-AAQB and Lomustine had no synergistic effect.We also found that 4-AAQB inhibited LPS-induced production of nitric oxide by monocytes. Taken together, results of this study suggest that the purified bioactive compound, 4-AAQB, from A. cinnamomea mycelium has the potential to inhibit the growth of meningeal carcinoma.
