蛹蟲草(Cordyceps militaris)菌絲體萃取物具有改善小鼠肺纖維化之潛力

Potential Effect of Cordyceps militaris Mycelium Against Bleomycin-induced Pulmonary Fibrosis in Mice

摘要


博來黴素(Bleomycin)為一種抗癌藥物,常應用於淋巴癌、睾丸癌等癌症治療上,然而副作用是造成肺部炎症及纖維化的情形發生。肺部纖維化若持續其病程,將可能造成患者咳嗽、低血氧甚至呼吸困難等狀況。本研究利用博來黴素進行誘導肺纖維化,以評估蛹蟲草菌絲體萃取物是否具有預防保護效果。本試驗分成三組,分別為健康對照組(healthy control; HC)、博來黴素組(BLM)及實驗組(CM),每組6隻C57BL/6小鼠,實驗組每日餵食一次蛹蟲草萃取物500 mg/kg持續14天,於第15天餵食完後,將小鼠以氣管注射方式投予3mg/kg bleomycin sulfate,再餵食小鼠試驗樣品持續21天後進行犧牲,並收集支氣管肺泡灌洗 液(bronchoalveolar lavage fluid, BALF)及肺組織樣本進行肺部病理分析。結果顯示,小鼠經餵食蛹蟲草菌絲體萃取物,可以顯著降低由博來黴素誘導肺纖維化組織中的膠原蛋白沉積及發炎細胞浸潤情形,說明蛹蟲草菌絲體萃取物具有減緩因博來黴素所造成之小鼠肺部纖維化潛力。

Bleomycin is an anticancer drug, which is often used in the treatment of cancers such as lymphoma and testicular cancer. However, the side effects are pulmonary inflammation and fibrosis. If pulmonary fibrosis persists, it may cause cough, hypoxemia, and even have difficulty in breathing. In this study, bleomycin was used to induce pulmonary fibrosis to evaluate whether the extract of Cordyceps militaris mycelium has preventive and protective effects. This experiment was divided into three groups, the healthy control (HC) group, the bleomycin group (BLM), and the experimental group (CM), with six C57BL/6 mice in each group. Mice in the experimental group were first fed with Cordyceps extract 500 mg/kg once a day for 14 days. On day 15, the mice were administered 3 mg/kg bleomycin sulfate by tracheal injection, and then the mice were fed with test samples until day 21. After sacrifice, the bronchoalveolar lavage fluid (BALF) and lung tissue samples were collected for lung pathological analysis. The results showed that feeding mice with Cordyceps militaris mycelium extract could significantly reduce collagen deposition and inflammatory cell infiltration in pulmonary fibrosis tissue induced by bleomycin. These results suggest that Cordyceps militaris mycelium extract has the potential to ameliorate bleomycin-induced pulmonary fibrosis in mice.
