豆腐廢水特性及其應用於乳酸菌培養之評估

Assessment of the Characterization in Tofu Wastewater and Application in Lactic Acid Bacteria Cultivation

摘要


本研究主要利用豆腐製作過程所產生之廢水(豆腐水,TFW)作為乳酸菌的培養基,並探討乳酸菌發酵豆腐水上清液(SCS)之抗氧化及抗菌活性。將植生型乳酸菌B0106及BCRC 10069培養於豆腐水中,其生長曲線於第20小時趨近於菌數穩定,可獲得菌數達9 log CFU/mL。添加1~4% glucose及2% peptone於豆腐水中,結果顯示B0106及BCRC10069培養後之菌數並無顯著增加。豆腐水之抗氧化力具有58.41 ± 0.1%清除DPPH自由基之能力、總多酚含量相當於沒食子酸323.01 ± 10.05ppm,γ-aminobutyric acid(GABA)含量為191.57 ± 6.77ppm;而乳酸菌B0106、BCRC10069發酵豆腐水分別具有59.89 ± 0.57%及60.30 ± 0.01%清除DPPH自由基之能力,總多酚含量則相當於沒食子酸339.36 ± 8.96ppm及307.27 ± 10.47ppm之含量,而GABA含量分別為189.76 ± 23.36ppm及228.82 ± 5.30ppm。進一步將乳酸菌發酵豆腐水應用於抗耐甲氧西林金黄色葡萄球菌(BCRC15211)之抑制,結果顯示B0106於第二小時即達到抑制BCRC 15211 50%的能力,且B0106與BCRC 10069分別於第六小時及第八小時達到抑制金黄色葡萄球菌BCRC 15211 90%以上之能力。因此豆腐水不僅含有豐富的營養成份與胜肽適合乳酸菌生長,乳酸菌發酵液亦具有良好的抗氧化活性及增加GABA含量之能力,同時也具有抑制抗藥性金黄色葡萄球菌生長的能力,將來應用於保健食品及化妝品則有著無限潛力。

The purpose of this study is to use the waste water (tofu waste water, TFW) obtained from the tofu production process as media for the cultivation of lactic acid bacteria (LAB). Then, antioxidative, antibacterial activity of the spent culture supernatant (SCS) of LAB were investigated. B0106 and BCRC 10069 strains were cultured in TFW, and their growth curve was stable at the 20th hour and the bacteria number was 9 log CFU/ mL. 1~4% glucose or 2% peptone were added into TFW, the results showed that B0106 and BCRC 10069 bacteria number did not increase significantly. The ability of TFW to scavenge DPPH free radicals was 58.41 ± 0.1%, the total phenolics content was equal to gallic acid 323.01 ± 10.05 ppm and the content of γ-aminobutyric acid (GABA) was 191.57 ± 6.77 ppm. The SCS of B0106 and BCRC 10069 cultivated in TFW had the ability to scavenge DPPH free radicals to 59.89 ± 0.57% and 60.30 ± 0.01%, respectively. The total phenolics content were 339.36 ± 8.96 and 307.27 ± 10.47 ppm, GABA contents were 189.76 ± 23.36 and 228.82 ± 5.30 ppm, respectively. In addition, SCS of B0106 and BCRC 10069 were used to inhibit methicillin-resistant Staphylococcus aureus (BCRC 15211) growth, the results showed that B0106 achieved 50% inhibitory activity against BCRC 15211 at the second hour, and inhibited BCRC 15211 growth more than 90% after the sixth hour. From the above results, tofu water is rich in nutrients for the growth of B0106 and BCRC 10069, the SCS of B0106 and BCRC 10069 have good antioxidant activity, high GABA content and strong inhibitory activity against BCRC 15211 growth.
