以中子放射活性化分析法定量綠藻生活環細胞所含之錳與銅

Determination of Contents of Manganese and Copper during the Life Cycle of Chlorella Cells by Neutron Activation Analysis

摘要


應用綠藻同調培養技術及非破壞中子放射活性化分析方怯,定量絲藻生活環各階段細胞所含錳與銅量,並求得其變化情況。各約50毫克乾燥綠藻細胞試料與錳、銅標準試料同時放入清華原子爐,於熱中子通量約10^(12)中子/平方厘米/秒下各照射半小時及2小時,經放置冷却後將試料及標準試料放於接在256 channel波高分析器之二英吋厚井型碘化鈉內,以γ線譜分析法分析。對於錳則比較試料與標準試料0.85 Mev峯之放射能,對於銅則比較試料與標準銅試料0.51 Mev峯之放射能而定量。結果得含錳40~90 p. p. m.,銅60~160 p. p. m.。並知綠藻生活環各階段裏細胞對於錳與銅之攝取,以細胞成熟期較其生長期為多。

By using the technique of synchronous culture of chlorella and non-destructive neutron activation analysis method, the changes in content of manganese and copper during the algal life cycle of chlorella cells were investigated. About 50 mg. of dry chlorella cell sample was irradiated simultaneously with standards in the Tsing-Hua reactor at a thermal neutron flux of about 10^(12)n/cm^2. sec. for half or two hours. After cooling the sample was mounted in a 2 inches thick, well-type sodium iodide crystal attached to a 256 channel pulse height analyzer. Manganese and copper were determined by the gammer ray spectrometry. Comparing the peak heights at 0.85 Mev. of the sample with that of standard manganese and at 0.51 Mev. of the sample with that of standard copper. Manganese of 40-90 p. p. m. and copper of 60-160 p. p. m. were determined. It was found that the manganese and copper were assimilated more in the ripening stage than the growing stage during the life cycle of chlorella cells.
