臺灣肖楠正己烷萃取物中氫氧化鈉可溶之部份以磷酸處理,溶於磷酸的成份中所含tropolone類化合物之量很少,且甚難分離。今發現將之先經活性矽氧膠柱層分離可除去大部分非tropolone類化合物,然後用新設計的柱層分離法以經磷酸浸漬之纖維素粉為吸着劑反覆分離之。薄層色圖分析法證明有β-Thujaplicinol, β-Thujaplicin, γ-Thujaplicin三種已知物及三種未知之tropolone類化合物存在,其中β-Thujaplicin 與γ-Thujaplicin 為主成份。作者並單離得β-Thujaplicinol, γ-Thujaplicin 及結構尚未確定的一種tropolone類化合物與一種含有羥基,羰基的雙萜酸甲酯。
In the phosphoric acid-soluble part of n-hexane extract from Lebocedrus formosana Florin, only about one-fifth was tropolonic substances which were very difficult to separate from the mixture. A new separation method was then applied: the mixture was first filtered through activated silica gel to get rid of the non-tropolonic components, and then it was column-chromatographed over phosphoric acid impregnated cellulose powders. By using thin-layer chromatography, β-Thujaplicinol, β-Thujaplicin, andγ-Thujaplicin were identified and the three other tropolonic components were detected. β-Thujaplicin and γ-Thujaplicin are the chief tropolonic components while others exist in small amounts, β-Thujaplicinol and γ-Thujaplicin were isolated, and another unidentified tropolone was separated as copper complex, m. p. 265~7°C (decompd.) A non-tropolonic substance, named Compound G, C_(21)H_(30-32)O_4, m. p. 140-1°C, was also isolated. From spectral data it could be proposed as a methyl ester of hydroxyketoacid in diterpene series.
