副腎皮質賀爾蒙及蛋白同化促進Steroid製劑之極譜定量法

Polarographic Assay for the Preparations of Corticoid and Protein Anabolic Steroids

摘要


Dexamethasone, methandrostenolone, durabolin等製劑以甲醇抽出,利用極譜法在50%甲醇,pH 6.3 之醋酸鹽緩衝液中,於E_(1/2)-1.24 V., -1.25 V., -1.41 v.分別測定該含量,所得到之結果相當準確,可適合於本類藥品之定量。在當做藥品使用的多數副腎皮質和荷爾蒙及蛋白同化促進steroids中,其極譜(polarogram)被研究的是屬於副腎皮質荷爾蒙的cortisone, hydrocortisone, prednisone, prednisolone 的分離定量,及關於這些極譜對pH濃度,溫度的影響,此外Robertson曾經檢討cortisone, hydrocortisone之極譜性質,而朝日則研究hydrocortisone, prednisolone 之極譜性質。由以上結果,已知道些steroids是屬於共軛系的Δ^4-3-keto steroids,且都表示出1電子1質子的還原波,羰基被還原成> COH,更會形成pynacol型化合物,其還原波高與水銀壓之平方根及濃度成正比,而有1.94%/℃之溫度係數,pH為零時的E_(1/2)是-1.03~-.07 v. vs. NCE, ΔE_(1/2)/ΔpH是-0. 059~0.063。然而對於其他最近合成的多種副腎皮質荷爾蒙及蛋白質同化促進steroids則尚無研究。因之,著者從事檢討dexamethasone, methandrostenolone, durabolin等之極譜的性質並研究其定量法。

Preparations of dexamethasone, ,methandrostenolone, and durabolin were extracted with methanol and determined polarographically in an actate buffer of pH 6.3 in 50% methanol at half-wave potential -1.24 , -1.25, and - 1.41 v. (S. C. E.), respectively. Both dexamethasone and methandrostenolone produce two reducing waves in the pH range between 2.8 and 10.2. At pH 6.3 the sum of the height of these two waves is proportional to the square root of mercury pressure and concentration within the range 0.2 to 1.8 mM. For dexamethasone the half-wave potential varies with the pH according to the equations E_(1/2) (1 st)= -0.84-0.063 pH and E_(1/2)(2nd)= -0.94-0.061 pH, while for methandrostenolone E_(1/2) (1 st) = -0.86-0.062 pH and E_(1/2) (2 nd) = -0.95-0.060 pH. Addition of gelatin causes the disappearance of the second wave. The remaining first wave is also proportional to the concentration. The diffusion current constant, l_d is about 1. Under the same condition, durabolin gives a reducing wave with El/2 at -1.41 v. The diffusion current is proportional to the concentration in the range from 0.2 to 1.8 mM. Its I_d value is found to be 1.1. The results of assay of standard mixture and commercial preparation were showed as follows: (The form abridges) The starch, lactose, talc, and magnesium stearate contained in the preparations do not interfer the results. Therefore, the polarographic "method is suitable for the assay of the preparations of Δ^(1,4)- 3 ketosteroids.
