以質譜技術為核心的蛋白質體研究

Mass spectrometry-based proteomics

摘要


近來,由於幾種重要的生物基因體被陸續地解碼,以及生物質譜儀器的日新月異,以質譜為主的技術平台已成為研究蛋白質體學的主力工具之一,首先,對蛋白質體學所造成的衝擊,即是質譜儀能對蛋白質進行快速的定性及定量分析.藉由分析蛋白質體在不同狀態下的表現差異,我們得以一窺細胞生理變化奧秘,也因為這樣,蛋白質前端分離準備工作,及這些分離工具與質譜儀的串聯對於蛋白質體學就顯得日益重要。本篇文章,我們將以質譜儀為出發點,並介紹其相關配套的蛋白質分離策略,亦將針對幾個應用價值的領域加以探討。

Recently, the complete sequences of several genome and advances of in instrumentation has surrender mass spectrometry a key role in proteomics-associated analysis. Protein identification and characterization by mass spectrometry-based methods have made a significant impact on proteomics, unraveling the cell physiology by profiling the differentially expressed proteins. Emerging strategies has allowed large-scale protein separation followed by precise characterization of the separated proteins. This review will highlight the advances in mass spectrometry and protein separation strategies relevant to proteomic research. Some fields where proteomics has provided valuable biological insight will also be discussed.
