免疫親和性層析的進展與應用

The development and application of immunoaffinity chromatography

摘要


利用抗體對抗原的專一性與親和性,將欲檢測之目標物的抗體固定於固態表面,並以此修飾之固態物填充管柱,作為液相層析儀的分析管,此種層析稱之為免疫親和性層析,因免疫親和性層析對檢測目標物及其類似物具有選擇性與親和性,所以能用於檢測處於極端複雜之環境中的目標物(如生物檢體),同時能夠分辨目標物與其類似物,相較於傳統層析方法,不但可以省去繁雜的樣品前處理流程,節省檢測時間,同時也節省濃縮與萃取所需的有機溶劑,對環境保護、操作人員的健康及檢測成本有極大的效益,本文將為大家介紹免疫親和性管柱的進展與應用。

By applying imunoaffinity column (IAC) as analytical column prepared by packing the material immobilized with the antibody of the analyte, immunoaffinity chromatography could extract and determine simultaneously the analyte and its analogs from the sample whose environment is extreme complicate due to the specific and affinity between the immobilized antibody and the analyte. Compared to the traditional chromatography, immunoaffinity chromatography has two advantages, one is it could avoid tedious procedures for sample pretreatments before injecting into HPLC system and this results in saving the labor for sample treatment. Another is it could avoid utilizing toxic organic solvent for sample extraction, and this avoids the impacts of the environment protection and the risk of the operator health. Based on these two advantages, immunoaffinity chromatography is a time-saving and safety method especial for biofluid sample determination. In this article, we will introduce the literatures in which IAC was used for sample determination and discuss the development and application of IAC.
