Establishment of High Throughput Screening (HTS) for Drug Discovery

摘要


高速篩選系統是一個很重要的核心技術,其應用在藥物開發上,可以篩選上千上萬個可以調節特定標的基因的化合物,特別的是此系統可以減少從開發藥物到上市的時間。近年來,親近閃爍檢測法,螢光共振能量轉移偵測,生物冷光分析,細胞毒性測試,偵測的孔盤模組和液體操作等新技術的改良,成功地增加篩選的速度和減少所需反應物的體積。此外,在高速篩選系統上,品質控制也是很重要的,Z'-factor提供一個很好的參數去鑑定分析系統的穩定性。這篇文章回顧幾種可執行於高速篩選技術平台的分析系統,這些分析系統將加速藥物的開法與發展。

High-throughput screening (HTS) is a core technology for drug discovery to screen hundreds of thousands to millions of compounds against specific molecular targets, specifically, to reduce the time-to-market in drug development. In recent years, advances in biotechnologies, such as sicintillation proximity assay (SPA), fluorescence energy transfer (FRET), bioluminescent assay, cell proliferation assay, detection platforms and liquid-handling technologies, have significantly increased throughput and reduced assay volumes. Furthermore, quality control (QC) is an important part in HTS. The Z’-factor provides a good parameter for evaluating assay performance. This review will summarize several approaches available for HTS formats that will speed up drug discovery and development.
