新穎體學質譜定量技術-SWATH

Novel Mass Spectrometry-Based Technique for Quantitative Omics-SWATH

摘要


隨著質譜儀的迅速發展,定量分析技術日新月異,sequential window acquisition of all theoretical fragment ion spectra (SWATH) 為2012 年由Ruedi Aebersold 團隊發表的一個新穎的串聯質譜數據收集模式。SWATH 利用非數據依賴擷取 (data independent acquisition, DIA) 模式能夠改善傳統的質譜數據依賴擷取 (data dependent acquisition, DDA) 模式,在有限樣品中擷取到最多的分析物訊號,並可從中獲得定量的資訊,得到與多重反應偵測 (multiple reaction monitoring, MRM) 媲美的定量結果。此種新穎質譜定量技術SWATH 分析方法可應用於多種體學研究,如蛋白質體學 (proteomics)、胜肽體學 (peptidomics)、脂質體學 (lipidomics)、代謝體學 (metabolomics),或是轉譯後修飾等,此技術可同時得到龐大的定性與定量數據,效率大幅超越傳統定量技術。相信在不久的未來,新穎質譜定量技術SWATH 將能夠為體學 (omics) 研究帶來新紀元。

With the rapid development of the mass spectrometry (MS), the technology of quantitative analysis hasmade great progress. In 2012, Ruedi Aebersold groupdeveloped a novel MS data acquisition mode-sequential window acquisition of all theoretical fragment ion spectra (SWATH). SWATH aims to complement traditional mass spectrometry-based qualification and quantitation techniques such as data dependent acquisition (DDA) and multiple reaction monitoring (MRM) methods. SWATH utilizes data independent acquisition (DIA) strategies to increase the reproducibility andcomprehensiveness of data collection andcan perform a larger number of MRM-like experiments concurrently. SWATH can be applied to various field ofthe Omics, such as proteomics, peptidomics, lipidomics, metabolomics, post-translational modification and etc.
