莨菪素抗氧化特性之評估研究

Evaluation and Study on the Antioxidant Characteristics of Scopoletin

摘要


莨菪素(Scopoletin)為枸杞、白芷等中藥之指標成份,是一種香豆素類的化合物。根據以往的研究報告指出,莨菪素具有許多種藥理作用,包括保護肝細胞、降血壓、抗菌、退燒、止痛等功用。以往曾有文獻指出莨菪素可能具備抗氧化活性,但對於莨菪素與抗氧化劑如維生素C或維生素E間之交互作用,則不曾有相關研究報告。本研究之主要目的在探討莨菪素與維生素C、維生素E是否具備有抗氧化之協同作用。我們分別測試莨菪素、維生素C、維生素E個別或相加之後,DPPH捕捉能力、總酚含量、ABTS+清除能力與還原力變化。實驗結果顯示,隨著莨菪素濃度愈高,其DPPH自由基捕捉能力,與維生素C或維生素E之協同的作用也愈明顯。在總酚含量測定之試驗中,莨菪素添加維生素C後之總酚含量也具有濃度愈高效果愈佳的情形;但在添加維生素E時,協同效果則不明顯。進一步的實驗發現莨菪素加入維生素C後,ABTS+清除能力與還原力也具有顯著的協同效果。後續研究將設計含有莨菪素、維生素C的抗氧化保養品配方,以細胞模式及人體試驗,評估其皮膚保養之有效性。

Scopoletin (6-methoxy-7-hydroxycoumarin) is a pharmacologically active coumarin compound that has been isolated from several traditional Chinese herbs, such as Lycium chinense Mill and Angelicae dahuricae Radix. Several studies have shown that scopoletin show hepatoprotective activity, antihypertension, anti-bacteriae, down fever and relieve pain. Previous study reported that scopoletin has potential antioxidation activity. However, there is no report about interaction between scopoletin and other antioxidants such as vitamin C or vitamin E. The aim of this research is to evaluate possible synergistic antioxidation effects between scopoletin and vitamin C or vitamin E. Antioxidation activity was assayed by determining the DPPH radical scavenging activity , total phenol content, ABTS+ radical cation decoloriation and reducing power. The experimental results showed that DPPH radical capture ability is higher along with scopoletin concentration. The capture ability is also more obvious when combined with vitamin C or vitamin E. When scopoletin was combined with vitamin C, the total phenol content also increased in a dosedependent pattern, whereas similar result was not found in the case of vitamin E. Additionally, scopoletin and vitamin C also showed remarkable synergistc effects on reducing power and the elimination of ABTS+. Further study will be performed to design cosmetic formulations containing scopoletin and vitamin C or other anti-oxidants, and the effectiveness of such skin care products will also be evaulated.
