莨菪素抑制酪胺酸酶功效之研究

Study on the Tyrosinase-Inhibiting Effects of Scopoletin

摘要


莨菪素(Scopoletin;6-methoxy-7-hydroxycoumarin)廣泛存在於植物中,可從芸香科白鮮屬、芸香科九里香屬等植物中萃取,為香豆素(coumarin)化合物之一。根據過去研究,莨菪素作用於家兔具有抗凝血的功效,此外,還具有抗菌、抗氧化、抗腫瘤等多項藥理活性。本研究初步先測試不同濃度莨菪素是否能抑制蕈菇酪胺酸酶活性,並將莨菪素與麴酸(Kojic acid)、酪胺酸酶一起反應,觀察酵素活性之變化情形。接著使用老鼠B16-F10黑色素瘤細胞來探討莨菪素對細胞內黑色素含量以及酪胺酸酶活性的影響。研究結果顯示,高濃度莨菪素(2 mM~10 mM)可以抑制蕈菇酪胺酸酶的活性,隨著濃度愈高,抑制效果愈佳。但與麴酸一起反應後的酵素抑制效果,與單獨使用莨菪素或麴酸比較,並沒有明顯的協同作用。而使用低濃度莨菪素(25 μM、50 μM、100 μM)能抑制細胞中酪胺酸酶活性,並且使B16-F10細胞內黑色素含量減少。未來將進一步探討莨菪素可能的美白機轉,並評估將莨菪素應用於美白保養品配方的可能性。

Scopoletin (6-methoxy-7-hydroxycoumarin)is one of coumarin compound that have been widely existed in the plant, such as Rutaceae Dictamnus、Rutaceae Murrayapaniculata. It was shown that scopoletin show the anti-hemoglutination effect on the domestic rabbit. In addition, scopoltin present many pharmacological activities, such as anti-bacterial, anti-oxidative and anti-tumor functions. The effects of scopoletin on mushroom tyrosinase activity, intracellular tyrosinase activity and melanin content of murine B16-F10 melanoma cells were evaluated. Kojic acid was used as a positive standard in the above experiments. The study showed that high concentration of scopoletin (2 mM~10 mM)may suppress mushrrom tyrosinase activity, in a dose-dependent pattern. However, when scopoletin combined with kojic acid, there is no obvious synergistic effect on the inhibition of tyrosinase activity. Interestingly, intracellular tyrosinase activity of B16-F10 cells was suppressed by low concentration of scopoletin (25 μM, 50 μM, 100μM). Besides, the melanin content in B16-F10 cells was also decreased by scopoletin in a dose-dependent pattern. The further work will elucidate the whitening mechanism of scopoltin, and we will also evaluate the possibility of scopoletin applied in the whitening cosmetic formulations.
