[6]-生薑醇抑制黑色素生成之研究

Inhibitory Effects of [6]-Gingerol on Melanogenesis

摘要


[6]-生薑醇由薑(Zingiber officinale Roscoe, Zingiberaceae)分離之一種辣味成分,以往的研究指出[6]-生薑醇具有抗氧化、抗血小板凝集、抗發炎、抗胃潰瘍、抑制血管新生、抗癌等藥理作用,但關於[6]-生薑醇對於黑色素合成之影響,則不曾有任何研究報告。本研究之主要目的,在於探討[6]-生薑醇是否具備抑制酪胺酸酶活性,降低黑色素之合成,進而表現出可能的美白活性。配製不同濃度的[6]-生薑醇與蕈菇類之酪胺酸酶反應,發現[6]-生薑醇(25 μM至100 μΜ)無法抑制細胞外酪胺酸酶酵素活性。然而以[6]-生薑醇處理B16F10老鼠黑色素瘤細胞後,發現[6]-生薑醇可以明顯抑制細胞中酪胺酸酶活性且減少黑色素合成量,其效果比麴酸(200 μM)佳。上述結果證實[6]-生薑醇能有效抑制黑色素瘤細胞之酪胺酸酶活性並且降低黑色素生成量,未來將設計含有[6]-生薑醇之化妝品配方,進行安全性及有效性評估,以進一步開發成為含有[6]-生薑醇之美白保養品。

[6]-Gingerol, is a pungent ingredient of ginger (Zingiber officinale Roscoe, Zingiberaceae). Previous reports showed [6]-Gingerol expresses various pharmacological activities such as anti-oxidation, antiplatelet aggregation, anti-inflammation, anti-ulcerogenesis, anti-angiogenesis and anti-carcinogenesis. However, the effects of [6]-Gingerol on melanogenesis have not been reported. This study was aimed to evaluate the potential whitening effects of [6]-Gingerol, including antimelanogenesis and/or inhibition of tyrosinase activity. The preliminary in vitro results showed that various concentrations (25-100 μΜ) of [6]-Gingerol do not surpress the catalytic activity of mushroom tyrosinase. Surprisingly, [6]-Gingerol inhibited intracellular tyrosinase activity and decreased melanin biosynthesis in B16F10 murine melanoma cells, whose effects are stronger than those of kojic acid (200 μΜ). In conclusion, [6]-Gingerol exerts antimelanogenic activity coupled with antityrosinase properties effectively. In the further study, [6]-Gingerol will be applied in cosmetic formulations. Also, effectiveness and safety of such products will also be assayed. It is predicted that [6]-Gingerol is useful in the skin whitening products.
