芳香製品中鄰苯二甲酸酯類的分析及其對人類角質細胞的毒性研究

Analysis of Phthalates in Aromatic Products and the Toxicity on HaCaT Cells

摘要


鄰苯二甲酸酯類(phthalates)廣泛的用於食物包裝、醫療及化妝品作為增塑劑、助溶劑或定香劑。苯二甲酸酯類屬於一種環境荷爾蒙,許多的報導指出其可能造成雄性老鼠生殖系統的毒性或產生肝毒性,但對於皮膚系統之傷害則較少著墨,本研究的目的即在研究市售之芳香製品包括香水、室內芳香劑及精油等產品中鄰苯二甲酸酯類的種類與含量分析,與對皮膚角質細胞的毒性作用。以氣相層析質譜儀(Gas Chromatography Mass)分析34種芳香製品後發現,在香水、室內芳香劑等20項產品中,11項產品(55%)含鄰苯二甲酸二乙酯(Diethyl phthalate, DEP),其DEP最高的劑量達19,800ppm。有1項產品(5%)含鄰苯二甲酸二丁酯(Dibutyl phthalate, DBP),其DBP的劑量高達36,085ppm。而在分析的14種精油中則並未發現有含有任何的鄰苯二甲酸酯類。在測量多種鄰苯二甲酸酯類對人類角質細胞株(HaCaT)的毒性後發現,DEP的IC_(50)為0.54mg/ml。我們的研究顯示不管國內或國外的產品,目前在香水中仍含有相當高比例的DEP或DBP,也提醒國人重視化妝品中含有鄰苯二甲酸酯類的安全性問題。

Phthalates are present in many daily consumer products, such as food wrappings、 medical devices and cosmetics as solvents or plasticizers. Phthalates are generally considered as environmental hormones. Many animal studies have shown that phthalates can damage male reproductive system. However, little is known about the phthalate on skin toxicity. In this study, we analyze 34 aromatics products by GC-MS and found that 11 samples (55%) and 1 sample (5%) contained diethyl phthalate (DEP) and dibutyl phthalate (DBP) respectively, in the number of 20 fragrances. None of 14 essential oils was detected with phthalates. In MTT cytotoxicity assay, DEP induced dose response toxicity on HaCaT cells ( IC_(50) : 0.54mg/ml). There is no significant cytotoxicity induced by DBP on HaCaT cells. This study strongly suggests us to reconsider the safety concern of phthalates used in cosmetics.
