黃岑苷與黃岑苷元美白功效之研究

Study on the whitening activity of the Chinese herb Scutellaria baicalensis Georgi

摘要


近年來化妝品市場朝向回歸自然的方向發展,宣稱天然而溫和的成分已成為市場主流。而尋找具有減少黑色素合成功能之中藥美白成分,成為研究人員的主要題材。以往研究報告指出黃芩具有多項藥理活性: 黃芩萃取液具有清除自由基及抗氧化能力;黃芩苷元在試管試驗中可抑制皮膚上的真菌及細菌生長;黃芩苷具有保護肝臟的作用與抗UV引起膜脂質的過氧化作用;但黃芩實際上對於皮膚美白的可能功效到目前為止則未有明確的研究報告。美白課題為東方女性所注重,本研究在探討黃芩苷與黃芩苷元對胞外酪胺酸酶活性及黑色素細胞瘤中黑色素含量的影響,以評估應用於美白產品開發的潛力。酪胺酸酶活性評估結果顯示黃芩苷的抑制效果較黃芩苷元顯著,50 μM與100 μM黃芩苷之酪胺酸酶活性抑制率為19.46 %與30.62 %;10 μM,100 μM與200 μM黃芩苷元之酪胺酸酶活性抑制率抑制率為7.20 %,10.87 %與12.90 %;。此外,以5 μM、10 μM及25 μM黃芩苷對於B_(16)F_(10)細胞的黑色素含量抑制率分別為0.27 %、1.33 %、4.27 %;而1 μM、3 μM及5 μM黃芩苷元對於B16F10細胞的黑色素含量抑制率分別為4.33 %、10.33%、6.33 %。結果顯示兩者於美白產品之開發均具潛力。

Skin care products contain Chinese herb have become more popular in recent cosmetic market. One major trend of research and development of skin whitening cosmetics is to screen or search natural and gentle skin whitening ingredients. It is reported that Scutellaria baicalensis Georgi express various pharmacological activities as follows: crude extracts of Scutellaria baicalensis Georgi present free radicalscavenaging and anti-oxidation capacity; in vitro experiments show that baicalein inhibit proliferation of fungi and bacteria; baicalin is reported to present liver protection function and decrease UV-induced membrane lipid per-oxidation. However, there is little report of the potential skin whitening effects of Scutellaria baicalensis Georgi. The study aim to evaluate the potential skin whitening effects of baicalin and baicalein. We determined their inhibitory effects on enzymatic activity of mushroom tyrosinase and B16F10 intracellular melanin content. Preliminary results indicated that the inhibition percentage of mushroom tyrosinase activity of 50 and 100 μM baicalin were 19.46 % and 30.62 % respectively. The inhibition percentage of mushroom tyrosinase activity of 10, 100 and 200μM baicalein were 7.20 %, 10.87 % and 12.90 % respectively. Baicalin show better effects than baicalein. On the other hand, the decrease percentage of B16F10 intracellular melanin content of 5, 10 and 25 μM baicalin were 0.27 %, 1.33 % and 4.27 % respectively. In addition, 1, 3 and 5 μM baicalein decreased B_(16)F_(10) intracellular melanin content by the percentage of 4.33 %, 10.33 % and 6.33 % respectively. The above results indicated that both baicalin and baicalein may be added in the skin whitening cosmetic formulation as active ingredients.
