以微射流對撞高壓均質法製備奈米結構脂質載體之製程探討

摘要


本研究是以微射流對撞高壓均質法製備含仿生脂質的NLC探討其製備條件及特性,以做為未來包覆化妝品活性物的載體。NLC之脂基質是由仿生脂質為固脂與液脂辛酸/癸酸甘油三酯混合而成。採用熱高壓均質法製備NLC,探討不同製程壓力及處理回數兩個參數對NLC平均粒徑及分佈(多分散性指數;PdI)的影響。研究結果顯示最佳化的製備條件為1000 bar壓力下處理2個回數。所得到NLC的平均粒徑及分佈(PdI)分別為131.48±13.28 nm和0.17±0.02。90天後粒徑測量及光學顯微鏡觀察結果無較大顆粒及聚集現象(aggregation)產生。可知本研究以最佳條件製備之仿生脂質NLC可得到最小粒徑及分佈並具良好的物理穩定性,顯示其可做為包覆化妝品活性物的良好載體。

The objective of this study was the preparation and characterization of bionic nanostructured lipid carriers (NLC) prepared by the jet- stream high pressure homogenization as potential carrier for cosmetics. The lipid matrix of NLC was using blends of bionic lipids and caprylic/ capric triglycerides. The effects of the preparation conditions on the mean particle size, polydispersity index (PdI) were investigated. Optimized NLC were produced by 1000 bar and 2 cycles. The obtained NLC assumed with the average particle size of 131.48±13.28 nm and the PdI of 0.17±0.02. During the storage time at both 5℃ and room temperature, the NLC dispersion remained stable, only a small negligible increase in particle size was noticed after 90 days. The observation of light microscopy shows absence of large particles or aggregates. Based on the high stability and the narrow distribution of particle size and PdI, the prepared bionic lipid NLC could be a novel dermal nanocarrier for cosmetic actives.
