原花青素B2抑制過氧化氫誘發人類內皮細胞氧化傷害

Procyanidin B2 Inhibits the Oxidative Damage Induced by Hydrogen Peroxide in Human Endothelial Cells

摘要


研究顯示,活性氧(Reactive oxygen species, ROS)是引起心血管疾病的重要因子之一。流行病學研究亦顯示富含黃酮類的飲食可以降低心血管疾病(Cardiovascular disease)的風險及併發症的發生。原花青素B2 (Procyanidine B2)為一種黃酮類(Flavonoid)化合物,常見於各種水果、可可及紅葡萄酒中,具有抗氧化(Anti-oxidation)及抗發炎(Anti-inflammatory)的功效。然而,目前原花青素B2對於內皮細胞氧化傷害的保護作用研究尚少。因此,在本研究中擬探討原花青素B2對於H_2O_2誘發人類EA.hy926內皮細胞氧化傷害的影響,我們測量DNA股斷裂、脂質過氧化(Lipid peroxidation)以及細胞內ROS的含量。結果發現原花青素B2可顯著抑制H_2O_2誘發EA.hy926內皮細胞的DNA損傷和脂質過氧化,並具有劑量關係。進一步以流式細胞儀分析發現原花青素B2也可顯著降低細胞內ROS含量。因此,我們推測原花青素B2可顯著抑制EA.hy926內皮細胞之DNA傷害及脂質過氧化,其中的可能機制是透過清除細胞內ROS。這些研究結果顯示原花青素B2具有預防心腦血管疾病的潛力。另外,原花青素B2是否與可藉由增加體內抗氧化酵素的活性而降低細胞內ROS的濃度,其相關機制是值得進一步探討的。

Many studies have shown that reactive oxygen species (ROS) are important causes of cardiovascular diseases. Epidemiological studies demonstrate the possible protective effects of flavonoid-rich foods against cardiovascular diseases and the associated comorbidities. Procyanidine B2 (PB2) is a flavonoid found in cocoa, fruits and red wine. A number of studies showed that PB2 may possess antioxidant and anti-inflammatory properties. However, little is known about the protective effect of PB2 on oxidative damages in endothelial cells. Thus, in the present study, we investigated the effects of PB2 on H_2O_2-induced DNA damage and lipid peroxidation in human EA.hy926 endothelial hybrid cells. We determined DNA strand breaks, lipid peroxidation, and the levels of intracellular ROS. The results showed that PB2 significantly inhibited H_2O_2-induced DNA damage and lipid peroxidation in a dose-dependent manner in EA.hy926 cells. Accordantly, PB2 also significantly decreased intracellular ROS levels. Overall, the data suggest that PB2 prevents DNA damage and lipid peroxidation via reducing the level of intracellular ROS in EA.hy926 cells exposed to H_2O_2.
