α及β-澱粉酶之共價層析及鹽析層析

Covalent Chromatography and Salting Out Chromatography of α and β-Amylases

摘要


利用epichlorohydrin(ECH)為偶合劑,由β-環狀糊精製備不溶性β-環狀糊聚合物。將Phenylmercuric acetate(有機汞)或octylamine(疏水性基)鍵結於β-環狀糊精聚合物, 製備兩種親和吸著劑。甘藷β-澱粉酶可吸著於鍵結有機汞之聚合物,且可以半胱胺酸溶離吸著之β-澱粉酶。黃麴菌及豬胰臟α-澱粉酶於鍵結疏水基之聚合物進行鹽析層析後,均被吸附,然可以0.1 M Iris-HCI, pH 8.0溶離。

Water-insolubleβ-cycloclextrin polymer was prepared by crosslinkingβ cyclodextrin (β-CD) with epichlorohydrin. Two affinity adsorbents were prepared by coupling phenylmercuric acetate or octylamine on theβ-CD polymer. Sweet potatoβ-amylase was adsorbed on the organomercuric. β-CD polymer and desorbed with cystein. α-amylase from Aspergillus oryzae and hog pancreas were retained on the cetylβ-CD polymer by salting out chromatography. However, the retained a -amylase were eluted with 0.1 M Tris-HCl buffer, pH 8.0.
